| 摘 要： | 探讨质粒重组体构建中，重组体连接反应的时间对转化与基因克隆结果的影响。方法，Ｃ ＥＢＶＢＨＲ１基因与ＰＢＶ２２１载体的妆为研究对象，对连反应在３ｈ、６ｈ、１２、２４ｈ４个时段连接液的转化效果进行研究。

| Abstract: | Objective:To study the influence of time on ligation in construction of recombinant plasmid.Methods:In the model of ligation between EBV BHRF 1 gene and vector pBV221,target genes and vectors were ligated for 3 6 12 and 24 hours,the results of transformation were analysed.Results:No bacteria grew in plates which were ligated for 3 and 24 hours.Only a few colonies(1.7) grew in plates which were ligated for 12 hours.A large amount of colonies(19) grew in plates which were ligated for 6 hours.By identification,9/10 colonies were positive containing recombinant plasmid.Conclusions:In the construction of recombinant plasmid,the time appropriate for ligation is 6 hours. |
