摘    要:目的 研究HTK液和UW液对体外培养的大鼠原代心肌细胞冷保存中的保护作用。方法 体外培养大鼠原代心肌细胞,分别用生理盐水,UW液和HTK(液在0℃-4℃保存心肌细胞6h、8h、10h、12h。检测保存液的AST和LDH-L,台盼蓝染色法计算心肌细胞的生存率。结果 UW液组和HTK液组的AST和LDH-L均比生理盐水组极显著低下,心肌细胞的生存率则极显著的高于生理盐水组。而在12h时,HTK液组的AST和LDH-L显著低于UW液组,心肌细胞的生存率在8h和10h也明显高于UW液组。结论 HTK液和UW液均对体外培养的大鼠原代心肌细胞的冷保存有明显的保护作用。而HTK液相对于UW液更能延长对心肌细胞的冷保存时间。

Abstract:[Objective] To study the protective effect of HTK solution and UW solution on rat cardiac muscle cells. [Methods] Using the model of primary culture of rats cardiac muscle cells, Saline, HTK solution and UW solution were reapplied to rat cardiac muscle cells at 0~4 ℃.After 6 h, 8 h, 10 h and 12 h, rat cardiac muscle cells impairment was evaluated via the livability of cells and by measuring AST and LDH-L in every preservation solution. [Results] The levels of AST and LDH-L were found to be significantly higher after 12 h preservation in UW solution group compared with HTK solution group. The livability of cells was found to be significantly lower after 8 h and 10 h preservation in UW solution group compared with HTK solution group. [Conclusions] The effect of HTK solution and UW solution is observably in prevention of rat cardiac muscle cells. HTK solution can prolong the time of preserving rat cardiac muscle cells than UW solution.
