甲醛对小鼠血液中抗氧化酶及微量元素影响

摘    要:目的:探讨甲醛对小鼠血液中抗氧化酶及微量元素的影响。方法:选取80只健康昆明小鼠(由北华大学实验动物中心提供),体重18~24g,雌雄各半。将小鼠随机分4组,即对照组、低剂量组(1/40LD50)、中剂量组(1/20LD50)、高剂量组(1/10LD50),每组20只,每日定时静式吸取2h,连续6周,对照组不接触甲醛,其他处置均与感染组相同:6周后小鼠断头取血1mL,与微波消解器中加入纯硝酸消解2min,定容至10mL,原子吸收法测定Zn、Mn、Fe含量,同时取出0.02mL全血,配成1mL溶血液,采用南京建成生物制品厂提供的测试盒测定GSH-Px活性、MDA的含量和总抗氧化能力。结果:甲醛染毒小鼠全血脂质过氧化变化分析中,与对照组比较,高剂量组小鼠GSH-Px的活性显著降低,中、高剂量组小鼠总抗氧化能力显著降低,而中、高剂量组MDA含量显著升高(P〈0.05);甲醛染毒全血微量元素Zn、Mn、Fe的变化分析中,与对照组比较,染毒6周中、高剂量组小鼠全血微量元素Zn、Fe明显降低(P〈0.05),高剂量组Mn含量明显降低(P〈0.05)。结论:甲醛可损伤抗氧化酶系统,使GSH-Px、总抗氧能力降低,MDA含量升高,全血微量元素Fe、Zn和Mn也显著降低,为探讨甲醛对机体健康影响的作用机制提供的线索。

Abstract:Objective:To explore of Formaldehyde Exposure on antioxidant enzymes and trace elements in blood of mice.Methods:Select 80 healthy Kunming mice(from the Beihua University Laboratory Animal Center),weight 18~24g,half male and half female.The mice were randomly divided into four groups,control group,low dose group(1/40LD50),medium doses group(1/20LD50)and high dose group(1/10LD50).Every group has 20 mice,daily inhalation static timing two hours for 6 weeks,no contact with formaldehyde in the control group but other disposition remained same as other experimental groups.After 6 weeks,mice were decapitated.Took blood 1ml and put in the microwave digestion device,adding pure nitric acid digesting for 2 minutes,keeping volume as 10ml,using atomic absorption spectrometry method to determine Zn,Mn,and Fe content.At the same time took the 0.02ml of the whole blood,dubbed 1ml solvent blood,using the test case from Nanjing Jiancheng Biochemical Factory to test GSH-Px activity,MDA content and total antioxidant capacity.Results:Mice exposed to formaldehyde-wide changes in lipid peroxidation analysis,compared with the control group,the activity in high-dose group GSH-Px was significantly reduced,the antioxidant capacity of middle and high-dose groups were both significantly decreased,while the high-dose group MDA levels were significantly increased(P0.05);for formaldehyde exposure of whole blood trace elements Zn,Mn,and Fe-change analysis,compared with the control group within 6w exposure,the whole blood trace elements,Zn and Fe,in high dose group decreased significantly(P0.05),Mn content in high dose group significantly declined(P0.05).Conclusion:The formaldehyde can damage the antioxidant enzyme system,so that GSH-Px,reduced the total antioxidant capacity,MDA content increased,whole blood trace elements Fe,Zn,and Mn were significantly reduced,which provided clues in exploring the health effects of formaldehyde on the body's mechanism.
