蛋白激酶C在鼻息肉中的表达及意义

摘    要:目的探讨蛋白激酶C在鼻息肉组织中的表达及其与嗜酸粒细胞凋亡的相关性。方法13例鼻息肉患者给予丙酸氟替卡松喷鼻治疗,采用流式细胞术检测激素治疗前后鼻息肉组织中蛋白激酶C表达及凋亡的嗜酸粒细胞,并与7例健康正常人下鼻甲黏膜进行比较。结果蛋白激酶C主要表达在嗜酸粒细胞,其次在血管内皮细胞、腺体周围的平滑肌细胞;13例鼻息肉组织中嗜酸粒细胞表面均有蛋白激酶C表达(100%),而7例健康下鼻甲黏膜中3例(42.86%)有蛋白激酶C表达,差异有显著意义(P<0.05)。糖皮质激素治疗后的鼻息肉组织中有蛋白激酶C表达的嗜酸粒细胞减少,与激素治疗前相比,差异有显著意义(P<0.05)。结论蛋白激酶C在鼻息肉组织中呈高表达,糖皮质激素可以有效抑制鼻息肉中蛋白激酶C的表达,减少嗜酸粒细胞的浸润。

Abstract:Objective To investigate the relationship between the expression of protein kinase C (PKC) and the apoptosis of eosinophlis in tissues of rhinopolypus.Methods 13 patients with rhinopolypus were treated by fluticasone. The expression of PKC and the apoptosis of eosinophlis before and after treatment were detected by flow cytometry, and the normal membrane of nosepiece in 7 healthy subjects was used as control.Results PKC expressed in cytoplasm in most eosinophlis, and expressed in part of vascular endothelial cells and smooth muscle cells. The positive rate of expression of PKC in tissues of rhinopolypus (100%) was significantly higher than that in normal nosepiece tissues (42.86%)(P<0.05).The positive expression rate of PKC in tissues of rhinopolypus was decreased after treatment,compared with that before treatment,there was a significant difference (P<0.05).Conclusion The expression of PKC is increased in most eosinophlis cells of rhinopolypus tissues. Treatment with fluticasone could depress the PKC expression in eosinophlis cell and decrease the infiltration of eosinophlis cells.
