不同途径摄取激素对大鼠骨密度、血清指标的影响

摘    要: 目的 观察肌肉注射、口服给予强的松的方法对大鼠骨密度、血清指标的影响。 方 法将45只雄性SD大鼠随机分为3组 (正常组15只、口服组15只、肌肉注射组15只),其中正常组大鼠作为阴性对照,不给予任何药物干预;口服组大鼠给予强的松 10 mg/(kg· d)灌胃;肌肉注射组大鼠给予强的松10 mg/(kg· 2 d)。观察给药12周后大鼠椎骨骨密度水平、血清钙、磷、碱性磷酸 酶的变化。 结果12周后,与正常组相比,口服组、肌肉注射组大鼠椎骨骨密度、血钙水平均显著下降(P<0.05),碱性磷酸酶、血 磷水平均显著升高。此外,肌肉注射组与口服组大鼠椎骨骨密度水平相比下降(P<0.05),血清钙、磷、碱性磷酸酶水平相比则无 明显差异(P>0.05)。 结论与正常组相比较,口服和肌肉注射强的松都能降低大鼠椎骨的骨密度值,同时对大鼠血清钙、磷、碱性 磷酸酶有一定的影响,该结论可为需长期使用激素的骨质疏松症患者临床安全合理用药提供相关实验依据。

Effect of metacortandracin administration method and time on bone mineral density in rats

Abstract: ObjectiveTo observe the effect of metacortandracin administration method and time on bone metabolism in rats. Methods A total of 45 SPF rats were randomly divided into three groups (Normal group Intragastric administration group Injection group). Normal group, as a control group, administrated with nothing, Intragastric administration group with prednisone 10 mg/(kg · d). Injection group with prednisone 10 mg/(kg · 2d) , which were observed 12 weeks. All rat s were examined for bone mineral density (BMD), serum calcium,phosphorus, alkaline phosphatase. Resultsafter 12th weeks, compared with Normal group, BMD and serum calcium decreased significantly while the phosphorus, alkaline phosphatase increased obviously in Intragastric administration group and Injection group (P<0.05). Compared with Intragastric administration group, BMD decreased significantly in Injection group (P<0.05) while the level of serum calcium, phosphorus, alkaline phosphatase were not changed significantly (P>0.05). ConclusionCompared with Normal group, both Intragastric administration and injection with prednisone could influence the level of BMD, serum calcium, phosphorus, alkaline phosphatase, while injection with prednisone could decrease the BMD more significantly, the research result shows some meaningful theroptic methods for the op patients.
