HPLC-TOF/MS快速鉴别白狼毒药材中的化学成分

摘    要:目的 采用高效液相色谱飞行时间质谱(HPLC-TOF/MS)对大戟科白狼毒2种药材狼毒大戟和月腺大戟中的化学成分进行快速鉴别.方法 色谱柱:安捷伦Zorbax Eclipse XDB C18(3.0mm×50 mm,1.8 μm),流动相为乙腈-水,梯度洗脱,流速0.4 mL·min-1,柱温20℃,紫外检测波长210 nm,进样体积5μL;飞行时间质谱采用ESI源,正离子模式监测,质量数扫描范围50~2 000.结果 从狼毒大戟提取物中得到23个稳定的色谱峰,月腺大戟提取物中得到26个稳定的色谱峰,并对其所代表的化学成分进行定性分析,发现狼毒大戟和月腺大戟中的化学成分有明显差异.结论 采用HPLC-TOF/MS可以快速鉴别大戟科2种狼毒药材中的化学成分,为狼毒药材的质量控制研究及临床合理应用提供依据.

Abstract:Objective To establish rapid HPLC-TOF/MS method to analyze the chemical constituents in Bailangdu including Euphorbia fischeriana Steud and Euphorbia ebiacteolata Hayata.Methods An Agi1ent Zorbax Eclipse XDB-C18 column(3.0 mm×50 mm,1.8 μm) was used for rapid separation and identification of chemical components.The mobile phase consisted of water-acetonitrile in the gradient elution,the flow rate was 0.4 mL·min-1,the column temperature was 25 ℃,the UV detection wavelength was 210 nm and the injection vo1ume was 5 μL.Time-of-flight mass spectrometer(TOF/MS) and electrospray ion source(ESI)were used in qualitative analysis under the positive ion mode,and mass scan range was m/z 50-2 000.Results A total of 26 peaks in the extract of Euphorbia fischeriana Steud and 23 peaks in the extract of Euphorbia ebiacteolata Hayata with good UV absorption and MS response on the chromatogram were selected and identified.The difference in the chemical compositions in the extract of Euphorbia fischeriana Steud and Euphorbia ebiacteolata Hayata was obvious.Conclusion HPLC-TOF/MS method can be used to determine the characterization of the chemical constituents in Bailangdu,which provides an evidence for the quality control and clinical application of Bailangdu.
