全新丹参素衍生物抗心肌细胞凋亡作用研究

摘    要:目的 研究全新丹参素衍生物ADTM对心肌细胞凋亡的影响.方法 体外培养H9c2心肌细胞,t-BHP诱导H9c2细胞凋亡,Hoechst染色观察ADTM对心肌细胞凋亡的影响;结扎大鼠左冠状动脉前降支建立大鼠心肌缺血模型,缺血24 h后提取心肌组织蛋白,Western blot方法检测心肌组织凋亡相关蛋白Bcl-2、Bax以及caspase-3的表达.结果 ADTM能显著抑制t-BHP诱导的H9c2心肌细胞凋亡;在大鼠心肌缺血模型中,明显提高Bcl-2蛋白的表达,降低Bax和caspase-3蛋白的表达,减少缺血心肌细胞凋亡.结论 ADTM可显著抑制心肌细胞凋亡,从而减轻心肌细胞缺血损伤.

Abstract:Objective To investigate the in vitro and in vivo anti-apoptosis effect of ADTM,a novel derivative of Danshensu,on cardiomyocytes.Methods Hoechst 33258 staining was used to detect H9c2 cell apoptosis induced by t-BHP in vitro experiment.A rat model of acute myocardial ischemia was established by the ligation of left coronary artery anterior descending branch,and the expression of apoptosis-related protein Bcl-2,Bax and caspase-3 was measured by Western blot.Results ADTM significantly inhibited the H9c2 cell apoptosis.ADTM up-regulated the expression of Bcl-2 protein and down-regulated the expression of Bax and caspase-3 in a dose-dependent manner.The biological activity of ADTM was remarkably better than Danshensu.Conclusion Inhibition of cardiomyocytes apoptosis may be a potential protective mechanism of ADTM against cardial ischemia injury.
