蓝心草颗粒的定性定量方法研究

摘    要:目的 建立蓝心草颗粒的定性定量方法.方法 对蓝心草颗粒中板蓝根、穿心莲、鱼腥草进行薄层色谱鉴别.用HPLC法测定穿心莲内酯、脱水穿心莲内酯的含量,采用Dikma DiamonsiL C18柱,以甲醇-水(48∶52)为流动相,流速为1.0 mL·min -1,检测波长为240 nm.结果 薄层色谱斑点清晰,阴性样品无干扰.穿心莲内酯进样量在0.036 6~1.464μg与峰面积线性关系良好,r=0.999 2,平均回收率为97.6%,RSD为1.3%(n=9);脱水穿心莲内酯进样量在0.041 9~1.677 μg与峰面积线性关系良好,r=0.999 9,平均回收率为98.9%,RSD为1.7%(n=9).结论 本法专属性强,准确度高,重现性好,可作为该产品质量控制方法.

Abstract:Objective To establish a method for the quality control of Lanxincao Granule.Methods Isatidis Radix,Andrographis Herba,and Houttuyniae Herba were authenticated by thin-layer chromatography.An HPLC method for the determination of andrographolide and dehydrographolide in Lanxincao Granule was established.A Dikma Diamonsil C18 column was used with methanolwater(48 : 52) as the mobile phase,the flow rate was 1.0 mL · min-1 and the detection wavelength was 240 nm.Results The spots on the TLC plate were clear.The negative sample was not disturbed.Good linearity was obtained at 0.036 6-1.464 μg(r=0.999 2) for andrographolide and 0.041 92-1.676 8 μg(r=0.999 9) for dehydrographolide.The average recovery was 97.6% (RSD=1.3%,n=9) for andrographolide and 98.9%(RSD=1.7%,n=9) for dehydrographolide.Conclusion The method is specific,accurate and reproducible,and it can be used for the quality control of Lanxincao Granule.
