摘    要:目的:研究黄芪超滤膜提取物对人SGC7901胃癌细胞增殖和转移的影响。方法:应用四氮唑蓝比色法(MTT法)测定黄芪超滤膜提取物对人SGC7901胃癌细胞增殖的抑制作用,应用半定量逆转录-聚合酶链反应(RT-PCR)法、Western-Blot法研究黄芪超滤膜提取物对SGC7901胃癌细胞环氧化酶(COX-2)、血管内皮生长因子-c(VEGF-c)表达的影响。结果:黄芪超滤膜提取物对SGC7901胃癌细胞的增殖有一定的抑制作用,使SGC7901胃癌细胞阻滞于G0/G1期,对SGC7901胃癌细胞COX-2、VEGF-c的表达有显著抑制作用。结论:黄芪超滤膜提取物能有效抑制SGC7901胃癌细胞的增殖周期,并可抑制COX-2、VEGF的表达。

Effects of the ultra-filtration extract from astragalus membranaceous on proliferation and metastasis of SGC7901 cells

Abstract:Objective:To study the effects of the ultra-filtration extract from astragalus membranaceous(UEAM) on proliferation and metastasis of SGC7901 cells.Methods:SGC7901 cells proliferation were detected by Tetrazolium assay(MTT) colorimetry.Cell cycle was analyzed by cytometry.Semiquantity reverse translation-polymerase chain reac-tion(RT-PCR) and Western blot were performed to detect the expression of cyclooxygenase-2(COX-2) and vascular endothelial growth factor-c(VEGF-c) in SGC7901 cells.Results:UEAM can inhibit proliferation of SGC7901 cells and arrest SGC7901 cells in G0/G1 period.There were obvious effects on inhibiting expression of COX-2 mRNA and VEGF-c mRNA of SGC7901 cells.Conclusion:UEAM can inhibit proliferation cell cycle,expression of COX-2 mRNA and VEGF-c mRNA of SGC7901 cells.
