血清LncRNA TapSAKI对肾结石患者术后并发症的预测价值

目的探讨血清长链非编码RNA(LncRNA)TapSAKI对肾结石患者术后并发症的预测价值。方法收集2020年5月至2021年4月辽健集团铁煤总医院收治的103例肾结石患者为研究对象,根据术后患者是否出现并发症将其分为并发症组(n=28)和无并发症组(n=75),并于同期随机选取60例健康体检者为对照组。采用实时荧光定量PCR法(qRT-PCR)检测病例组术前及对照组血清LncRNA TapSAKI水平,酶联免疫吸附法检测血清肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)、单核细胞趋化蛋白-1(MCP-1)水平。采用Pearson积矩相关分析血清LncRNA TapSAKI与TNF-α、IL-6、IL-1β、MCP-1的相关性,绘制受试者工作特征(ROC)曲线评估血清LncRNA TapSAKI对术后并发症的预测价值。结果肾结石患者术后14 d内并发症发病率为27.18%(23/103)。并发症组、无并发症组血清LncRNA TapSAKI及TNF-α、IL-6、IL-1β、MCP-1水平高于对照组,且并发症组高于无并发症组(P<0.05)。经Pearson积矩相关分析,肾结石患者血清LncRNA TapSAKI与TNF-α、IL-6、IL-1β、MCP-1均呈正相关性(r=0.537、0.622、0.596、0.608,P<0.05)。ROC曲线结果显示,血清LncRNA TapSAKI预测术后并发症的AUC为0.835,临界值为1.92,灵敏度、特异性分别为0.87、0.82。结论血清LncRNA TapSAKI表达上调是肾结石患者术后并发症独立危险因素,早期检测可作为预测术后并发症的指标。

Predictive value of serum LncRNA TapSAKI for postoperative complications in patients with renal calculi

Objective To investigate the predictive value of serum long chain non-coding RNA(LncRNA)TapSAKI for postoperative complications in patients with renal calculi.Methods A total of 103 patients with renal calculi admitted to Tiemei General Hospital of Liaojian Group from May 2020 to April 2021 were selected as the research objects.According to the postoperative complications,patients were divided into complication group(n=28)and non-complication group(n=75),and 60 healthy subjects were randomly selected as the control group during the same period.Quantitative real-time polymerase chain reaction(qRT-PCR)was used to detect serum LncRNA TapSAKI levels in the case groups before operation and the control group.Serum levels of tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),interleukin-1β(IL-1β)and monocyte chemotactic protein-1(MCP-1)were determined by Enzyme-linked immunosorbent assay.Pearson product-moment correlation was used to analyze the correlation between serum LncRNA TapSAKI and TNF-α,IL-6,IL-1βand MCP-1,and receiver operating characteristic(ROC)curve was drawn to evaluate the predictive value of serum LncRNA TapSAKI in postoperative complications.Results The incidence of complications in patients with renal calculi within 14 days after operation was 27.18%(23/103).The levels of serum LncRNA TapSAKI,TNF-α,IL-6,IL-1βand McP-1 in the complication group and the non-complication group were higher than those in the control group,and those in the complication group were higher than those in the non-complication group(P<0.05).Pearson product-moment correlation analysis showed that serum LncRNA TapSAKI was positively correlated with TNF-α,IL-6,IL-1βand MCP-1 in patients with renal calculi(r=0.537,0.622,0.596,0.608,P<0.05).ROC curve results showed that serum LncRNA TapSAKI predicted postoperative complications with AUC of 0.835,critical value of 1.92,sensitivity and specificity of 0.87 and 0.82,respectively.Conclusion The up-regulation of serum LncRNA TapSAKI expression is an independent risk factor for postoperative complications in patients with renal calculi,and early detection can be used as an indicator to predict postoperative complications.
