侧脑室微量注射神经生长因子对实验性抑郁症大鼠抑郁行为和海马神经元损伤的影响

摘    要:目的:观察中枢给予外源性NGF对实验性抑郁症大鼠行为和海马神经元损伤的影响。方法:采用长期不可预见性中等强度应激造成大鼠抑郁症模型,运用侧脑室微量注射技术、Openfield行为测定、组织化学、光镜和是镜技术观察中枢给予外源性NGF对实验性抑郁症大鼠抑制行为和海马神经元损伤的影响。结果:侧脑室微量注射NGF可显著改善实验性抑郁症大鼠的抑郁行为,使实验性抑郁症大鼠海马CA1、CA3及齿状回(DG)神经元数目显著增加,酸性磷酸酶片面性显著降低,明显改善海马神经元的超微结构,结论:外源性给予NGF可能通过减轻大鼠海马神经元的损伤,从而改善动物的抑郁行为。

Abstract:Objective To observe the effects of i.c.v. NGF on the depressive behavior and changes of the hippocampal neurons in the depressive rats. Methods The depression model of rats was established by chronic unpredictable mild stress. The techniques of intracerebralventricular microinjection, openfield testing, histochemistry, light microscope and electron microscope were used to observe the effects of i.c.v. NGF on the depressive behavior and changes of the morphological and number of hippocampal neurons in the depressive rats. Results Compared with the i.c.v. saline , i.c.v. NGF could improve the depressive behavior significantly, mitigate the decrease of number of hippocampal neurons, decrease the activity of ACP in hippocampal neurons and the morphological changes of hippocampal neurons were ameliorated. Conclusion I.c.v. NGF might improve depressive behavior by protecting the hippocampal neurons from the injuries induced by experimental depression.
