Abstract:Objective Screen the clinically significant long-chain non-coding RNA(LncRNA)in cervical cancer,and study the biological function and mechanism of this LncRNA in the occurrence and development of cervical cancer.Methods The clinical database GEPIA was used to find genes that are highly expressed in cervical cancer,and the clinically significant LncRNAs were screened based on the clinical prognosis correlation(the principle of high expression and poor prognosis);the expression of this LncRNA was knocked down through siRNA-mediated,cell proliferation experiments and clone formation experiments were used to verify the importance of this LncRNA in the occurrence and development of cervical cancer;explored the nuclear and cytoplasmic location of this LncRNA,and finally analyzed the signal pathways involved in the co-expression of this LncRNA to explore the function of this LncRNA in cervical cancer potential molecular mechanism.Results A total of 78 highly expressed genes with significant clinical prognosis in cervical cancer were screened through the GEPIA database.There were 3 LncRNAs.DLEU1 was significantly highly expressed in cervical cancer(P<0.05),and patients with high expression had poor prognosis(P=0.0023);knocking down the expression of DLEU1 in cervical cancer cells by two siRNAs(P<0.001、P=0.001),significantly inhibited the proliferation of cervical cancer cells(P<0.05)and clone formation;DLEU1 is located in the cell nucleus,the expression of MYC,a regulatory oncogenic factor,was significantly lower(P<0.001、P=0.002).Conclusion DLEU1 may positively regulate the high expression of MYC through high expression,thereby promoting the growth of cervical cancer cells.
