糖尿病肾病患者血清miRNA-93的表达变化及意义

目的探讨miRNA-93在糖尿病肾病患者血清的表达情况及其临床意义。方法用实时荧光定量PCR技术分别检测20例糖尿病肾病患者血清和健康对照组血清中miRNA-93的表达变化。分析其与临床病理参数的关系;运用TargetScan等靶基因预测分析软件预测miRNA-93靶基因。结果与健康对照组相比,糖尿病肾病患者血清中miRNA-93低表达(P<0.05);尿毒症组患者血清中miRNA-93较非尿毒症组低表达(P<0.05);生物信息学分析显示为糖尿病肾病中miRNA-93的靶基因为VEGFA、MTHFR、ADRA2B、SDC2、APOB、PPARG。结论 miRNA-93在糖尿病肾病患者血清中低表达,可能通过调控其下游靶基因在糖尿病肾病发生过程中起重要作用。

Expression and significance of serum miRNA-93 in diabetic nephropathy patients

Objective To study serum microRNA-93(miRNA-93) expression in diabetic nephropathy patients and its clinical significance.Methods The expression levels of serum miRNA-93 in diabetic nephropathy patients and healthy controls were measured by quantitative real-time polymerase chain reaction(qRT-PCR).The relationship between miRNA-93 and the clinicopathological parameters in diabetic nephropathy was investigated.Target genes of miRNA-93 were predicted with bioinformatics softwares.Results Compared to healthy controls,serum miRNA-93 was significantly decreased in diabetic nephropathy patients(P<0.05).In addition,serum miRNA-93 was also significantly decreased in uremic patients compared with non-uremic patients(P<0.05).The potential target genes of miRNA-93 identified by bioinformatics softwares include VEGFA,MTHFR,ADRA2B,SDC2,APOB and PPARG.Conclusion Serum miRNA-93 is low-expressed in diabetic nephropathy patients.It may play an important role in the development and progression of diabetic nephropathy through its target genes.
