摘 要: | 目的探索一种成功率高、操作简便的肺纤维化大鼠模型制备方法。方法将36只雄性Wistar大鼠随机均分为对照组和实验组,对照组用手术暴露气管、肉眼直视下针刺滴入博来霉素(BLM)的方法造模,实验组采用经改良的气管插管注药方法造模。两组人员分别操作,各进行9只后交换人员进行剩余大鼠的操作。观察造模时间、各造模组大鼠的现场死亡率、后续死亡率、并发症及术者操作体会;第14天、28天分批处死大鼠,比较两组大鼠HE染色肺组织的病理情况。结果两种造模方法均能使大鼠肺组织产生典型的肺泡炎或纤维化改变,大鼠死亡率差异无统计学意义,均未见明显并发症;实验组操作时间显著低于对照组(P0.05),插管操作还具有操作上手度高、所需器材少、对麻醉要求低的优点。结论经改良的经气管插管注药方法较针刺注药操作时间短、所需器材少、操作简便,值得推广应用。

Abstract: | Objective Toexplorethepreparationofpulmonaryfibrosisratmodelwithasimpleand highsuccessfulratemethod.Methods Thirty-sixmaleWistarratswererandomizedintoacontrolgroup and an experimental group.In the control group,the model was prepared by infusion of Bleomycin(BLM)with the trachea exposed in surgery.In the experimental group,the model was prepared by the modified endotra-cheal intubation with medicinal infusion.The special persons were in the two groups and exchanged after modeling of every 9 rats.The modeling time,on-spot mortality of rats group,follow-up mortality and com-plications were observed in each modeling group as well as the experiences of the operators.The rats were sacrificed on the 14th and 28th days separately.The pathological condition of lung tissue was compared with HEstainingbetweenthetwogroups.Results Withthemodelingmethods,thetypicalpulmonaryalveolitisor fibrosis was induced in lung tissue of rats in the two groups.The mortality was not different significantly be-tween the two groups and the apparent complication was not found.In the experimental group,the operation time was less significantly than that in the control group(P〈0.05 ).The intubation operation was advanta-geousatsimplemanipulation,lessmaterialsandanesthesiarequired.Conclusion Themodifiedendotracheal intubation with medicinal infusion is characterized as short duration of operation,less materials required and simple manipulation.Hence,it deserves to be promoted. |
