药用真菌白桦茸液体深层发酵条件优化的研究

摘    要:目的研究采用液体发酵方法培养药用真菌白桦茸时,各种条件对白桦茸生物量形成的影响。方法采用碳源、氮源单因子实验及Plackett-Burman实验设计,筛选出最重要的几个因素,用minitab统计分析软件建立多元二次回归模型程并利用方差分析实验结果,并对最优条件做验证实验。结果在利用液体发酵法培养白桦茸时,酵母膏和装液量是对白桦茸液体培养生物量生成最重要的影响因素,培养的最适条件为温度为27℃,培养基中葡萄糖2%、磷酸二氢钾0.1%、硫酸镁0.05%、VB20.001%、氯化钙0.01%,酵母膏0.3%,装液量150 mL/250 mL,摇床转速140 r·min-1,在此条件下,实测细胞干重收率最高达到12.227 g·L-1。结论优化结果准确可靠,适用于白桦茸的液体发酵培养。

Abstract:Objective To study the impacts of every condition on biomass formation of Inonotus obliquus during culturing medicinal fungi Inonotus obliquus with submerged fermentation.Methods The carbon source and nitrogen source single factor experiment and Placket-Burman experiment design were adopted to screen the most important factors.The minitab statistical analytic software was used to set up the multiple quadratic regression model and ANOVA was applied to the experimental results.The optimum conditions were tested for verification.Results In the submerged fermentation of Inonotus obliquus,yeast extract and liquid filling amount were the most important factors for the biomass formation in submerged culture.The most suitable conditions of culture:27 ℃C,glucose 2%,KH2 PO4 0.1%,MgSO4 0.05 %,VB2 0.001%,CaCl2 0.01% in medium,yeast extract 0.3%,liquid filling amount 150 mL/250 mL and revolving speed 140 r · min^-1.In those conditions,the yield of dry cell weight measured actually was up to 12.222 7g · L^-1.Conclusion The optimized results are accurate,reliable and suitable for submerged fermentation culture of Inonotus obliquus.
