窄头橐吾抗癌活性成分的研究

摘    要:目的研究窄头橐吾的抗癌活性成分。方法采用柱层析(CC)对窄头橐吾根部的提取物进行分离提纯,并利用现代波谱方法(1H-NMR,13CNMR,HMBC,MS,IR)分析鉴定,确定其化合物的结构,用SRB方法,对从植物中分离而得的化合物进行抗癌活性实验。结果从该植物提取物的乙酸乙酯部分分离出3个化合物,其结构分别鉴定为:2,2-二甲基-4-(5,6-二甲氧基-2-苯并呋喃基)-4-戊烯酸(2,4-二甲氧基-5-醛基)苯酯(化合物1),1β,6α-二羟基-4(15)-桉叶烯(化合物2),2-异丙烯基-5-乙酰基-6-羟基苯并呋喃(化合物3),其中化合物1和3具有较好的抗癌活性。结论化合物1~3均为首次从该植物中分离而得,化合物1和3可能为潜在的抗癌药物。

Abstract:Objective To study the antitumor activity constituents from the roots of Ligularia stenocephala.Methods The compounds were isolated by means of silica gel column chromatography(CC) and elucidated by spectroscopic experiments.The antitumor activity assay was carried out according to the SRB method.Results Three compounds were obtained and established as(2,4-dimethoxy-5-al)phenyl 2,2-dimethyl-4-(5,6-dimethoxy-2-benzofuranyl)-4-pentenate(1),1β,6α-dihydroxy 4(15)-eudesmene(2),5-acetyl-6-hydroxy-2-isopropenylbenzofuran(3),compounds 1 and 3 showed appreciable antitumor activity against Bel-7402 and HO-8910.Conclusion Compounds 1~3 were all isolated from this plant for the first time,compounds 1 and 3 are potential antitumor drugs.
