健脑方促进脑缺血后大鼠海马神经发生的作用研究

目的观察中药健脑方对脑缺血后大鼠海马齿状回神经发生以及空间学习记忆功能的影响。方法SD大鼠随机分为假手术组、模型组和治疗组。采取永久结扎双侧颈总动脉制作全脑缺血模型。术后治疗组给予健脑方灌胃干预,模型组给予等量生理盐水。使用BrdU标记技术观察缺血后海马齿状回的神经发生,采用Morris水迷宫试验评价大鼠空间学习能力。结果模型组和治疗组海马齿状回BrdU阳性细胞数分别在缺血后第7天达到最高峰。而缺血后7、14、20、28 d治疗组BrdU阳性细胞数明显多于模型组(P<0.05或P<0.01)。水迷宫试验显示术后第14天治疗组大鼠寻台平均时间明显短于模型组(P<0.05)。结论健脑方能持续明显促进脑缺血后大鼠海马齿状回神经发生,并改善大鼠的空间学习能力。

Jiannao Recipe promotes neurogenesis of hippocampus in rats with cerebral ischemia

Objective To explore the effect of Jiannao Recipe on neurogenesis in the dentate gyrus and spatial learning and memory of rats with cerebral ischemia.Methods SD rats were randomly divided into sham group,model group and treatment group.The cerebral ischemia model was established by clamping the bilateral common carotid arteries permanently.Rats in the treatment group were orally administrated with Jiannao Recipe after operation,the model group with NS.Bromodeoxyuridine(BrdU) was used to label proliferating cells in the dentate gyrus.Spatial learning ability was tested by using Morris water maze test.Results BrdU positive cells in the dentate gyrus of the model group and the treatment group all reached the peak at day 7 after ischemia.BrdU Positive cells of the treatment group were obviously more than those in the model group at day 7,14,20 and 28 after ischemia(P<0.05orP<0.01).Morris water maze test displayed the average escape latency of the treatment group was obviously shorter than that of model group at day 14 after ischemia(P<0.05).Conclusions Jiannao Recipe can promote neurogenesis in the dentate gyrus of rats with cerebral ischemia continually,and improve spatial learning ability.
