卵巢癌组织中uPA与PAI-1 mRNA表达及意义

摘    要:目的 探讨尿激酶型纤溶酶原激活剂(uPA)和纤溶酶原激活剂抑制物-1(PAI—1)mRNA在卵巢癌组织中的表达及其临床意义。方法 采用逆转录-聚合酶链反应技术,检测38例卵巢癌、21例卵巢良性上皮性肿瘤及17例正常卵巢组织中uPA及PAI-1 mRNA的表达,并分析其与病理分化程度的相关性。结果 uPA及PAI—1 mRNA在卵巢癌中的表达水平均较卵巢良性上皮性肿瘤、正常卵巢组织显著升高(F=184.51、51.00,q=11.34~26.69,P〈0.05)。在不同分化程度的卵巢癌组织中uPA及PAI-1 mRNA的表达量随肿瘤分化程度的降低而增加(F=110.62、38.43,q=3.10~6.86,P〈0.05)。结论 卵巢癌组织中uPA及PAI—1 mRNA呈高表达,且与卵巢癌的病理分化程度相关。

Abstract:Objective To explore the expressions and their significance of urokinase-type plasminogen activator(uPA) and plasminogen activator inhibitor-1(PAI-1) in ovarian cancer. Methods The expressions of uPA mRNA and PAI-1 mRNA were evaluated by RT-PCR in the tissues of ovarian cancer(n=38),epithelial benign tumor(n=21) and normal ovary(n=17). Results The expressions of uPA mRNA and PAI-1 mRNA in the cancer tissue were higher than that in the benign tumor and normal ovarian tissue(F=184.51,51.00;q=11.34-26.69;P<0.05).A negative correlation existed between the expressions of uPA mRNA and PAI-1 mRNA and the differentiation of the cancer,the higher of expressions of uPA mRNA and PAI-1 mRNA,the lower differentiation of ovarian cancer were observed(F=110.62,38.43;q=3.10-6.86;P<0.05). Conclusion uPA mRNA and PAI-1 mRNA were over-expressed in ovarian cancer,and negatively correlated with the pathologic differentiation.
