大鼠脑出血后血肿周围TIMP-1的表达及与脑水肿的关系

摘    要:目的观察大鼠脑出血后血肿周围组织基质金属蛋白酶组织抑制剂-1(TIMP-1)的表达,探讨TIMP-1与脑出血后脑水肿的的关系。方法采用自体血注入法建立大鼠脑出血模型,分别用Western blot方法检测血肿周围组织的TIMP-1的表达,伊文思蓝(EB)测血脑屏障(BBB)通透性,干湿重法测组织含水量,并与对照组相比较。结果血肿周围组织的组织含水量和EB含量均48 h达到高峰,且二者呈正相关。TIMP-1在脑出血后12~36 h表达减低,且在脑出血后1~24 h时TIMP-1的表达与组织含水量以及EB含量都呈负相关。结论大鼠脑出血后血肿周围组织存在TIMP-1的表达下调,其可能促进了脑出血后早期脑水肿形成。

The expression of TIMP-1 in perihematoma and relationship of TIMP-1 to brain edema formation after intracerebral hemorrhage in rats
Abstract:Objective To investigate the expression of TIMP-1 in perihematoma and relationship of TIMP-1 to brain edema formation after intracerebral hemorrhage(ICH) in rats.Methods ICH models of rats were established by autologous blood injection.Then the expressions of TIMP-1 was detected through Western blot,the permeability of blood-brain barrier(BBB) was evaluated by Evans blue contents and the brain water content was measured by comparing wet and dry weight in perihematoma.Resules The brain water content and Evans blue contents in perihematoma were all increased and reached the peak at 48 h after ICH.There was positive correlation between them.The expression of TIMP-1 in perihematoma decreased at 12-36h after ICH.TIMP-1 was negatively correlated to the brain water content or Evans blue contents at 1-24h after ICH.Conclusion The expression of TIMP-1 in perihematoma was decreased after ICH in rats,which could be correlated with the brain edema formation at the early of ICH.
