健脾化痰法对抑郁模型大鼠海马内环境的影响

目的]研究海马内环境稳态在抑郁症中的作用。方法]40只雌性Wistar大鼠分为对照组、模型组、蒸馏水组和中药组。ELISA法检测血清皮质醇、雌二醇,光镜和电镜观察海马CA3区形态学变化,免疫组化法检测海马脑源性神经生长因子和血管内皮生长因子表达。结果]与对照组比较,模型组和蒸馏水组皮质醇水平显著升高,雌二醇水平明显下降,海马CA3区神经元损伤严重,脑源性神经生长因子和血管内皮生长因子表达明显降低,中药组上述指标变化不明显。结论]抑郁状态下,海马内环境稳态被破坏,定志小丸具有一定的维持海马内环境稳态的作用。

Effect of Vigoriting Spleen and Expelling Phlegm Method on Change of Hippocampal Internal Enviroment in Depression-mode Rats

Objective]To study the action of hippocampal internal enviroment in deprssion.Methods]40 femal wistar rats were divided into the control,model group,distilled water group and Chinese medicine group.Serum cortisol and estradiol were measured with ELISA.Hippocampal CA3 morphology were observed by light and electron miroscope.BDNF and VEGF expressions were detected by immunohistochemical assay.Results]Compared with those in the control,in the model group and the distilled water group,the serum cortisol level increased obviously,serum estradiol level decreased significantly,and the CA3 neurons had severious structure damage,and the expressions of BDNF and VEGF decreased markedly,while above index changed little in the Chinese medicine group.Conclusion]The homeostasis of hippocampal internal enviroment is disrupted in depression,and dingzhixiaowan can maintain this homeostasis.
