和胃接骨胶囊对实验性大鼠骨折愈合过程中BMP-2表达的影响

摘    要:为探讨和胃接骨胶囊对实验性大鼠骨折愈合过程中骨形态发生蛋白-2表达的影响,取SD大鼠60只,随机分为生理盐水组、麝香接骨丹组、和胃接骨胶囊组。徒手造成大鼠下肢胫腓骨标准骨折模型,造模成功后分别于1、2、4、6周每组断颈法处死5只大鼠,取骨痂组织制成病理切片,采用免疫组化染色技术进行染色,并用图像分析系统对骨形态发生蛋白-2的表达进行定量分析。结果显示,在骨折愈合过程中各组骨形态发生蛋白-2含量的总体变化趋势基本相同,骨形态发生蛋白-2表达的高峰都出现在骨折后第2周,但和胃接骨胶囊组骨形态发生蛋白-2阳性产物含量优于同期其他两组(P〈0.05)。表明和胃接骨胶囊能诱导骨形态发生蛋白-2的合成,可能是其促进骨折愈合的机理之一。

Abstract:In order to explore the effects of "HEWEIJIEGU JIAONANG"(HWJG,a TCD capsule for fracture healing) on expression of bony morphogenetic protein-2(BMP-2) in fracture rat,60 SD rat-models established by normative fracture method of tibia and fibula were divided randomly into 3 groups: Group A(20 rats,treated by physiological saline),Group B(20 rats,treated by "SHEXIANG JIEGU DAN"),Group C(20 rats,treated by HWJG);5 rats of each group were killed in four interval of 1,2,4 and 6 weeks postoperatively,the callus were excised,sliced,stained specimens by using the immunohistochemistry technology and analyzed BMP-2 expression with image analyses-system.The results showed that BMP-2 of each group was much the same in the overall change tendency,and expressive peak appeared the second week after fracture,but the BMP-2 masculine product of Group C was preponderate over that of the others(P<0.05),suggesting HEJG has a good function to improve the expression of BMP-2,and this maybe one of mechanism to promoting fracture healing.
