白介素-24抑制胶质瘤细胞生长的体内外实验研究

摘    要:目的探讨白介素-24基因转染对胶质瘤生长的影响。方法将载有人白介素-24eDNA的新型重组腺病毒(Ad5F35-IL24)感染人胶质瘤细胞系U251,体外观察Ad5F35-IL24对该细胞系生长的影响;建立人胶质瘤动物模型,瘤内注射Ad5F35-IL24,观察肿瘤生长情况;用流式细胞仪检测其凋亡和Bax/Bel-2的表达情况。结果Ad5F35-IL24感染U251细胞和肿瘤组织后,IL-24蛋白高表达,U251细胞和组织的生长受到明显抑制,其凋亡率明显升高,Bax/Bel-2也明显升高。结论白介素-24具有抑制人胶质瘤细胞系U251生长和诱导凋亡作用。Ad5P35-IL24转导白介素-24基因可能是一种有效的胶质瘤基因治疗途径。

Abstract:Objective To explore the potential use and the effect of adenovirus-mediated interleukin 24 gene(Ad5F35-IL24)therapy for malignant glioma.Methods After Ad5F35-IL24 was transfected into human malignant glioma cell line U251,we observed the growth of U251 cells.Ad5F35-IL24 was injected into tumor nodules of BALB/c mouse models of human glioma,and the tumor growth was observed.The apoptosis of tumor cells and their Bax/Bcl-2 expressions were detected with flow cytometer.Results The expression of IL-24 proteins were expressed highly after transfection of Ad5F35-IL24.The growth of U251 cells and xenograft tumors was suppressed obviously,and their apoptosis and Bax/Bcl-2 expression were significantly elevated.Conclusion IL-24 induces growth suppression and apoptosis in human glioma cells U251.Adenovirus-mediated IL-24 gene therapy is a potential effective approach in the treatment of malignant glioma.
