苦参碱诱导人乳腺癌细胞MCF-7/ADR的凋亡研究

摘    要:目的 研究苦参碱对人乳腺癌MCF—7/ADR细胞的生长抑制作用及诱导凋亡机制。方法 四甲基噻唑蓝(MTT)法测定苦参碱对细胞的半数抑制浓度(IC50),用荧光显微镜、电子显微镜、激光共聚焦显微镜、流式细胞仪和DNA凝胶电泳等技术观察细胞凋亡。结果 0.3—2.5g/L浓度苦参碱对MCF—7/ADR细胞有明显的抑制作用,并诱导发生凋亡,实验范围内细胞凋亡的比率与药物浓度呈一定相关。结论 苦参碱能抑制人乳腺癌MCF—7/ADR细胞的生长,其机制与阻止细胞周期的进程,启动细胞自身调控程序,诱导凋亡有关。

Abstract:Objective To investigate the growth-inhibition and the mechanism of apoptosis induced by matrine on MCF-7/ADR cell line.Methods Using MTT assay,the half-inhibition concentration (IC 50 ) of marine on MCF-7/ADR cells was detected.Fluorescence microscopy,electron microscopy,laser scanning confocal microscopy,flow cytometry (FCM) and DNA gel electrophoresis were applied to demonstrate the presence of apoptosis.Results Apoptosis was detectable in 0.3-2.5 g/L matrine-treated MCF-7/ADR cell line.The apoptosis-inducing effect of matrine was dose-dependent in the experiment range.Conclusion Matrine can induce the inhibition of MCF-7/ADR cell growth by apoptosis and the mechanism of apoptosis is related to the blocking of S period cells.
