长链非编码RNA XIST和miR-101-3p在子宫颈癌组织中的表达及关系

目的：探讨长链非编码RNA（LncRNA）XIST和miR-101-3p在子宫颈癌组织中的表达情况及其临床意义。方法：收集58例子宫颈癌和配对的癌旁组织标本，采用实时荧光定量PCR（qPCR）检测组织中LncRNA XIST和miR-101-3p的表达，分析其与子宫颈癌临床病理因素的相关性，并评价其临床意义。结果：与癌旁组织比较，qPCR显示宫颈癌组织中LncRNA XIST的表达明显上调（P < 0.05），miR-101-3p的表达明显下调（P < 0.05）；且二者在表达水平上呈负相关（r=-0.68，P=0.000）。LncRNA XIST和miR-101-3p的表达均与宫颈癌淋巴结转移、组织大小及临床分期之间存在相关性（P均<0.05）。结论：LncRNA XIST与miR-101-3p的表达呈负相关，可能成为早期筛选宫颈癌的生物标志物和治疗靶点。

Expressionof LncRNA XIST and miR-101-3p in cervical cancer

OBJECTIVE:To investigate the expression and clinical significance of long non-coding RNA (LncRNA) XIST and miR-101-3p in cervical cancer. METHODS:Cervical cancer tissues and matched non-tumor para-neoplastic tissues were collected from 58 patients. Quantitative real-time PCR was used to examine the expressionof LncRNA XIST and miR-101-3p,the correlation between them and their correlation with clinicopathological features were analyzed.RESULTS:LncRNA XIST expression was significantly up-regulated in cervical cancer tissues than in adjacent non-tumor tissues (P<0.05). The expression of miR-101-3p was significantly down-regulated in cervical cancer tissues as compared with the adjacent tissues (P<0.05).The expression of them in nucleic acid levels were negatively correlated(r=-0.68,P =0.000)and correlated significantly with FIGO stage,lymph node metastasis,and tissue size of tumor (P<0.05). CONCLUSION:Our findings indicate that LncRNA XIST and miR-101-3p may be potential prognostic markers and therapeutictargets for cervical cancer. The expression of LncRNA XIST and miR-101-3pwas negatively correlated.
