胃癌中微小RNA let-7i的低表达及其临床意义

目的:探讨微小RNA let-7i在胃癌中的表达及其与临床病理特征的关系。方法:用茎环RT-PCR的方法检测68例胃癌患者的肿瘤组织及癌旁正常组织中let-7i的表达,并分析其与胃癌临床病理指标及化疗敏感性的关系。结果:胃癌中let-7i的表达明显低于配对的癌旁正常组织(P<0.001),且let-7i的异常表达与肿瘤的局部侵袭(P<0.001)、淋巴结转移(P<0.006)、分化程度(P<0.018)及化疗抵抗(P<0.001)显著相关。结论:miRNA let-7i可能是一个潜在的生物标志物和分子靶标,可在胃癌的早期诊断和基因治疗中发挥作用。

Decreased expression of microRNA let-7i in gastric cancer and its clinical significance

Objective:To investigate the expression of miR let-7i in gastric cancer and its relationship with clinicopathologic features.Methods:The expression of let-7i in 68 pairs of primary gastric cancer and normal adjacent tissues(NATs) was quantified by stem-loop RT-PCR,and the relationship between let-7i level and clinicopathologic features and chemosensitivity was explored.Results:Let-7i was significantly down-regulated in cancer tissues compared with the paired NATs(P < 0.001).Low expression of let-7i showed significant correlation with local invasion(P < 0.001),lymphatic metastasis(P = 0.006),poor differentiation(P = 0.018),and resistance to chemotherapy(P < 0.001).Conclusions:These findings indicate that let-7i may be a potential biomarker and molecular target for early diagnosis and gene therapy in gastric cancer.
