摘 要: | 目的:比较三种赣产葛根及其地上部分中总黄酮的含量,采用正交试验得到葛地上部分总黄酮提取的最佳工艺条件。方法:以葛根素为对照品,采用紫外-可见分光光度法测定三种赣产葛根、茎、叶和花中总黄酮的含量,以乙醇浓度、料液比和提取时间为考察因素,通过正交法确定葛地上部分总黄酮的最佳提取工艺。结果:不同基源的葛及其地上部分总黄酮含量差异较大,经正交试验考察,得最佳提取工艺为:乙醇浓度60%、料液比1:50、回流提取1.5 h。结论:该含量测定方法操作简单、快速、准确,可用于葛不同部位总黄酮含量的同时测定。总黄酮提取工艺参数的优化将为葛地上部分资源的开发利用提供理论基础和科学依据。

Optimization of extraction technology and content determination of total flavonoids from aerial parts of three kinds of Pueraria lobate from Jiangxi Province |

Abstract: | Objective The content of total flavonoids in three kinds of P. lobata and its aerial parts from Jiangxi were compared, and the optimum extraction conditions of total flavonoids from aerial parts of P. lobata were obtained by orthogonal test. Methods The content of total flavonoids in the roots, stems, leaves and flowers of P. lobata from Jiangxi Province was determined by ultraviolet-visible spectrophotometry with Puerarin as the control substance. Considering the concentration of ethanol, the ratio of material to liquid and the extraction time as factors, the optimum extraction technology of total flavonoids from aerial parts of P. lobata was determined by orthogonal design. Results The content of total flavonoids in different origin of P. lobata and its aerial parts is quite different. The best extraction process parameters were obtained by orthogonal test: ethanol content 60%, solid to liquid ratio1:50(g/mL), extracting time 1.5 h. Conclusion The content determination method is simple, rapid and accurate, which can be used for the simultaneous determination of total flavonoids in different parts of P. lobate. Optimization of extraction process parameters of total flavonoids will provide theoretical basis and scientific basis for the development and utilization of the aerial part of P. lobata. |
