摘    要:目的:应用基因芯片技术,检测nm23-H1基因沉默后K562细胞中与肿瘤相关基因的差异表达情况,探讨nm23-H1基因沉默对K562细胞生物学行为变化的分子作用机制。方法:采用Trizol一步法抽提K562-siNM23细胞和K562-siNC细胞总RNA;用RNA扩增试剂盒进行双链cDNA的合成及荧光标记cRNA的合成,并纯化cRNA。与功能分类基因芯片杂交反应后,用计算机分析比较2种细胞中与肿瘤相关基因的差异性表达。结果:共筛选出表达差异性基因14个,其中表达上调基因9个(ratio2);表达下调基因5个(ratio0.5)。结论:nm23-H1基因沉默后,引起显著变化的基因与细胞周期调控和抗凋亡信号转导等生物学功能相关。
