HERG通道阻断剂对胃癌细胞增殖能力的影响

摘    要:目的探讨HERG通道阻断剂对胃癌细胞增殖能力的影响。方法应用HERG通道阻断剂干预胃癌细胞的HERG电流后,采用MTT法对胃癌细胞进行生长曲线的描绘;采用平板克隆形成实验检测胃癌细胞的克隆形成能力;采用流式细胞计数检测胃癌细胞细胞周期分布的变化。结果HERG通道阻断剂对胃癌细胞的增殖有抑制作用,而对GES细胞的增殖则无明显抑制作用;随着药物剂量的增加及作用时间延长,其抑制作用显著增强。HERG通道阻断剂降低了胃癌细胞的克隆形成能力(P〈0.05),阻止胃癌细胞由G1期进入S期。结论HERG通道阻断剂可抑制胃癌细胞的增殖和克隆形成能力,并抑制胃癌细胞G1/S期转化,说明HERG电流在胃癌细胞的增殖中发挥作用,HERG蛋白是一个潜在的胃癌生物治疗的靶分子。

Abstract:Objective To investigate the effect of HERG channel blocker on the proliferation of gastric cancer cells. Methods HERG channel blocker was used to interfere HERG current of gastric cancer cells, and then the growth curve of gastric cancer cells was drawn by MTT method;The ability of clone formation of gastric cancer cells was studied by clone formation assay;The cell cycle distribution of gastric cancer cells was investigated by flow cytometry. Results The proliferation of gastric cancer cells was inhibited in a time- and dose-dependent manner when gastric cancer cells were treated with HERG channel blocker(cisapride),but cisapride had little effect on proliferation of gastric epithelial cells. The clonogenicity of gastric cancer cells treated with cisapride decreased compared with control group (P 〈 0.05 ). Cisapride tended to inhibit gastric cancer cells entering S phase from G1 phase in the cell cycle. Conclusion HERG channel blocker can inhibit the proliferation, clone formation and progression of G1/S of gastric cancer cells, which indicates that HERG current may play a role in the proliferation of gastric cancer cells, and HERG protein is a potential target for gastric cancer biological therapy.
