加味归脾汤对乳腺癌细胞抑制作用的研究

摘    要:目的:研究加味归脾汤对人类乳腺癌细胞的抑制机制。方法:不同浓度的加味归脾汤作用于人类乳腺癌ER+代表株MCF-7和ER-代表株MDA-MB-435,阳性对照药为多西紫杉醇(Docetaxel),通过MTS、细胞凋亡分析和Caspase-8检测等方法评价各药物浓度组的抑制效果。结果:MCF-7和MDA-MB-435的加味归脾汤高浓度组细胞抑制率在48h时均高于其Docetaxel组(P0.05),72h时MCF-7较其Docetaxel组略低(P0.05),MDA-MB-435与其Docetaxel组相近。流式细胞仪72h检测结果显示MCF-7和MDA-MB-435的加味归脾汤高浓度组和Docetaxel组凋亡程度相近。加味归脾汤中、低浓度组效果低于Docetaxel组。Caspase-8检测显示MCF-7和MDA-MB-435的加味归脾汤高浓度组检测结果接近或超过Docetaxel组(P0.05),加味归脾汤中、低浓度组均低于Docetaxel组。结论:加味归脾汤高浓度药液对ER+、ER-乳腺癌细胞抑制效果均可达到或超过阳性对照药。初步确定Caspase途径为其机制之一。

Abstract:Objective:To study the inhibited effects and principles of Modified Spleen Decoction on breast cancer cells.Methods:The MCF-7(ER+)and MDA-MB-435(ER-)breast cancer cell strains were inhibited by Modified Spleen Decoction different concentration to evaluate their inhibited effects respectively and investigate its principles synthetically from the cells suppression ratio through MTS,the apoptosis analysis by flow cytometry and the results of Caspase-8 tests.The positive control medicine was Docetaxel.Results: The inhibited effect of Modified Spleen Decoction on MCF-7 and MDA-MB-435 were higher than the Docetaxel group at 48h(P<0.05),and the lower than Docetaxel group on MCF-7(P<0.05);identical on MDA-MB-435 at 72h.The Flow cytometry showed that the degree of apoptosis was similar between the Modified Spleen Decoction of high concentration and Docetaxel group.Meanwhile,the inhibited effect of the other Modified Spleen Decoction groups were worse than the Docetaxel group.The Caspase-8 showed that the Modified Spleen Decoction of high concentration was higher than the Docetaxel group at 48h and 72h.both on MCF-7 and MDA-MB-435(P<0.05).And the other concentration of Modified Spleen Decoction were lower than Docetaxel group.Conclusion: The inhibited effect of Modified Spleen Decoction of high concentration can meet or exceed the one of positive control medicine on either ER+or ER-.One of the passway is Caspase.
