蒙医温针对疲劳大鼠作用的神经内分泌机制研究

摘    要:目的探讨蒙医温针对疲劳大鼠的作用及神经内分泌机理。方法将SD雄性大鼠随机分为正常对照组、模型组和温针组,每组20只。采用21d力竭游泳实验制作运动性疲劳模型。取蒙医顶会和心穴,使用MLY-Ⅰ型蒙医疗术温针仪进行温针。每隔2d治疗1次,每次15min,共7次。在造模第21天观察大鼠体重、力竭游泳时间和悬尾不动时间,并在末次运动后24h取材。用高效液相-电化学法检测下丘脑去甲肾上腺素(NE),多巴胺(DA)及其代谢产物3,4-羟基苯乙酸(DOPAC)、高香草酸(HVA),5-羟色胺(5-HT)及其代谢产物5-羟吲哚乙酸(5-HIAA);检测血清和睾丸丙二醛(MDA)、谷胱甘肽(GSH)、谷胱甘肽过氧化物酶(GSH-PX)和超氧化物歧化酶(SOD);用ELISA方法测定血清皮质酮、睾酮;计算睾丸指数和肾上腺指数。结果温针治疗可以明显延长力竭游泳时间(P<0.05)、缩短悬尾不动时间(P<0.05),使下丘脑5-HIAA、HVA含量明显下降(P<0.05),使睾丸和血清GSH含量明显增加(P<0.05),睾丸SOD活性明显增加(P<0.05),血清皮质酮水平明显降低(P<0.01),睾酮/皮质酮比值明显增高(P<0.05)。各组体重无显著性差异(P>0.05),各组睾丸指数和肾上腺指数无显著性差异(P>0.05)。结论蒙医温针可以提高疲劳大鼠运动能力,改善疲劳状态,这可能与其调节中枢单胺类神经递质含量、提高机体抗氧自由基能力、抑制亢进的下丘脑-垂体-肾上腺轴有关。

Abstract:Objective To investigate the effects and neuro-endocrine mechanism of mild needle therapy of mongolian medicine on fatigue rats.Methods Male SD rats were evenly randomized into control,model and mild needle groups.The model was established by exhausted swim.Mild needle therapy of mongolian medicine was applied to Dinghui and Xinxue for 15 min,once every 2 days.On the 21st day of the model establishment,the exhausted swim time,the immobility time in tail suspension test were measured.24 h after the last swim,the rats were decapitated.The norepinephrine(NE),dopamine(DA),dihydroxy-phenyl acetic acid(DOPAC),homovanillic acid(HVA),5-hydroxytryptamine(5-HT),5-hydroxyindoleacetic acid(5-HIAA) in hypothalamus were detected by HPLC-ECD method.The malondialdehyde(MDA),glutathione(GSH),glutathione peroxidase(GSH-PX) and superoxide dismutase(SOD) in serum and testis were measured.The corticosterone and testosterone content in serum were detected by ELISA.The testis index and adrenal gland index were calculated.Results Mild needle therapy significantly prolonged the exhausted swim time(P<0.05),and reduced the immobility time in tail suspension test(P<0.05).It also lowered the 5-HIAA and HVA content in hypothalamus(P<0.05,0.05).It significantly increased GSH in serum and testitis(P<0.05,0.05) and increased SOD in testitis(P<0.05),and significantly reduced corticosterone(P<0.01) and improved T/C(P<0.01) in serum.There were no significant differences among 3 groups in body weight,testis index and adrenal gland index(P>0.05).Conclusion Mild needle therapy of mongolian medicine can increase the monoamine content in rat brain,and improve the activity of antioxidant substances in the body.It may also reduce corticosterone in serum,improve T/C,which suggest it may regulate the hyperactivity of HPA axis.So they may underlie the action of mild needle therapy in raising the exercise ability and improving fatigue.
