细胞增殖核抗原及Bcl-6在损伤脊髓组织中的表达意义

目的 探讨高压氧(HBO)对脊髓损伤(SCI)脊髓组织中细胞增殖核抗原(PCNA)和Bcl-6表达的影响。方法 复制SCI模型，采用免疫组织化学染色，观察各组脊髓组织中PCNA和Bcl-6的表达变化。结果 正常对照组脊髓组织结构完整，神经细胞形态正常；神经元及神经纤维中PCNA和Bcl-6均阴性。损伤组组织出血、水肿、空泡变、神经纤维排列紊乱、断裂，神经元减少；PCNA和Bcl-6强阳性表达于神经元细胞中，坏死灶内弥漫性强阳性表达。0．1MPa HBO组的HE及免疫组织化学染色结果相似于损伤组；而0．25MPa HBO组则组织水肿消失，出血、空泡变性减轻，神经细胞形态恢复，结构排列相对完整；PCNA和Bcl-6阳性表达于神经元细胞中，但表达强度明显低于单纯损伤组。结论 HBO治疗后脊髓组织的细胞由趋于凋亡向修复转变。

Significance of cellular proliferative nuclear antigen and Bcl-6 expression in spinal cord injury

Objective To study the effect of hyperbaric oxygen (HBO) on the expressions of cellular proliferative nuclear antigen (PCNA) and Bcl-6 in spinal cord injury (SCI).Methods The expression changes of PCNA and Bcl-6 were observed with immunohistochemical staining.Results In normal control group, the structure of spinal cord was integrated, the nervous cells normal. PCNA and Bcl-6 were all negative in neuron and nerve fibers. In the injury group, there were hemorrhage, edema, vacuolation in spinal cord, disorder and fragmentation in nerve fibers. PCNA and Bcl-6 all strongly positively expressed in neuron, and diffusely positively expressed in necrotic foci. The results of 0.1MPa HBO group was similar with that of injury group; while in 0.25MPa HBO group, the edema disappeared, the vacuolation eliminated, the form of neurous cells recovered, and the arrangement of structure relatively normal. The degree of PCNA and Bcl-6 expressions decreased in comparison with that of injury group.Conclusion The cells in spinal cord converse from apoptosis to recover after treated with HBO.
