乳腺癌中hsa-miR-24-3p的表达及临床意义

摘    要:目的观察hsa-miR-24-3p在乳腺癌组织中的表达及与乳腺癌临床病理特征的关系,探索其发挥作用的具体机制。方法采用组织原位杂交实验检测乳腺癌组织和乳腺良性病变组织中hsa-miR-24-3p的原位表达,针对112例乳腺癌临床病理参数和相应hsa-miR-24-3p的表达水平进行相关性分析;预测hsa-miR-24-3p下游靶基因,通过荧光素酶报告实验和实时荧光定量PCR实验进行验证。结果在乳腺癌组织和乳腺良性病变组织中hsa-miR-24-3p的阳性率分别为36.6%(41/112)和66.7%(14/21)。hsa-miR-24-3p表达与乳腺癌患者的腋窝淋巴结转移数以及肿瘤的TNM分期呈负相关(P <0.05),与患者年龄、肿瘤直径、组织学分级、ER、PR和HER-2的表达均无显著相关性(P >0.05)。hsa-miR-24-3p表达与乳腺癌患者的无复发生存期和总体生存期呈正相关(P =0.025,P =0.019)。hsa-miR-24-3p靶向抑制RNF2的表达。结论 hsa-miR-24-3p在乳腺癌中具有抑癌功能,其可作为乳腺癌的潜在靶向治疗药物。

Abstract:Purpose To observe the expression of hsa-miR-24-3p in breast cancer tissues and to explore its clinicopathologic significance and its specific mechanism. Methods The expression of hsa-miR-24-3p was detected by tissue in situ hybridization. The correlation between the clinicopathological parameters and the expression of hsa-miR-24-3p was analyzed in the 112 breast cancer patients. The downstream target genes of hsa-miR-24-3p were predicted and verified by luciferase reporter assay and qRT-PCR. Results The positive expression rates of hsa-miR-24-3p in breast cancer tissues and benign breast lesions were 36.6%(41/112) and 66.7%(14/21) respectively. The expression of hsa-miR-24-3p was negatively correlated with the number of axillary lymph node metastasis and TNM stage of breast cancer patients ( P <0.05),but was not related to age of the patient,the size of the tumor,the histological grade of the tumor,and the expression of ER,PR and HER-2 ( P >0.05). The expression of hsa-miR-24-3p was positively correlated with both relapse-free survival and overall survival of breast cancer patients ( P =0.025 and P =0.019). hsa-miR-24-3p directly targeted and suppressed the expression of RNF2. Conclusion hsa-miR-24-3p performed tumor suppressing role in breast cancer patients which suggest hsa-miR-24-3p can be used as a potential target for breast cancer therapy.
