摘    要:目的:建立肠杆菌科食源性感染常见致病菌的快速检测方法。方法:选带正电荷的尼龙膜作为寡核苷酸芯片载体,采用寡核苷酸芯片技术对肠杆菌科食源性感染常见致病菌进行检测和鉴定。结果:在同一条件下运用多重PCR扩增出14种(属)细菌的16SrRNA、23SrRNA基因片段,随后应用寡核苷酸芯片技术对致病菌进行检测得到了相应的特异性杂交图谱。结论:建立的寡核苷酸芯片技术可快速检测肠杆菌科食源性感染常见致病菌,为食源性感染的快速诊断与预防奠定了良好的基础。

Abstract:Objective:To construct a rapid and accurate detection method of Enterobacteriaceae foodborne infection bacteria with oligonucleotide microarray technique.Methods:Nylon membrane was used as oligonucleotide microarray base.An experimental procedure was set up applying oligonucleotide microarray technology to detect and identify Enterobacteriaceae foodborne infection bacteria.Results:The 16S rRNA and 23S rRNA gene fragments of 16 species of pathogenic bacteria were amplified with multi-PCR at the same condition.The corresponding specific hybridization maps were obtained applying oligonucleotide microarray technology to detect and identify Enterobacteriaceae foodborne infection bacteria.Conclusion:The oligonucleotide microarray technology can rapid detect and identify pathogenic bacteria,and provides a good way in the diagnosis,therapy and control of foodborne disease.
