苯那普利对糖尿病大鼠肾组织RAGE表达的影响

摘    要:目的通过研究苯那普利对糖尿病大鼠肾脏组织糖化终末产物受体(RAGE)表达的影响,探讨其肾保护机制。方法30只SD大鼠随机分为正常对照组(n=10)、糖尿病对照组(n=10)和糖尿病苯那普利处理组(n=10)。实验第12周末,分别计算各组大鼠肾质量/体质量比值;检测血糖、糖化血红蛋白(HbA1c)和血脂水平,并行24 h尿蛋白定量;运用RT-PCR法测定大鼠肾组织RAGE mRNA表达。结果与正常对照组比较,糖尿病对照组和糖尿病苯那普利处理组大鼠的血糖、HbA1c、甘油三酯、总胆固醇和低密度脂蛋白胆固醇均明显升高(P<0.05),高密度脂蛋白胆固醇明显降低(P<0.05),而糖尿病对照组与糖尿病苯那普利处理组之间上述指标比较无显著差异(P>0.05)。糖尿病苯那普利处理组大鼠的肾质量/体质量比值、24 h尿蛋白定量和肾组织RAGE mRNA表达明显高于正常对照组大鼠(P<0.05),但显著低于糖尿病对照组大鼠(P<0.05)。结论苯那普利可抑制糖尿病大鼠肾脏肥大并降低尿蛋白排泄率,其作用机制可能与抑制RAGE表达有关。

Abstract:Objective To investigate the effects of benazepril on the expression of receptors for advanced glycation end-products(RAGE)in diabetic rat kidneys,and to explore its mechanisms of renal protection in diabetic rats. Methods Thirty SD rats were randomly divided into three groups(n=10 in each group): normal control group,diabetic control group and diabetic with benazepril group.Blood glucose,blood lipid,HbA1c,kidney to body weight ratio and 24 h urinary protein excretion were detected after 12 weeks.RAGE mRNA level was analyzed by quantitative RT-PCR. Results Compared with the normal control group,the blood glucose,HbA1c,triglyceride,total cholesterol and low density lipoprotein cholesterol in the diabetic control group and diabetic with benazepril group were significantly increased(P<0.05),and high density lipoprotein cholesterol significantly decreased(P<0.05),while there were no such differences between the diabetic control group and diabetic with benazepril group(P>0.05).The kidney to body weight ratio,24 h urinary protein excretion and RAGE mRNA level in the diabetic with benazepril group were significantly higher than those in the normal control group(P<0.05),and lower than those in the diabetic control group(P<0.05). Conclusion Benazepril can prevent renal hypertrophy and decrease urinary protein excretion in diabetic rats.The mechanism may be related to its ability to inhibit RAGE expression.
