绞股蓝皂苷对人衰老皮肤成纤维细胞抗氧化酶活性的影响

摘    要:目的 在细胞水平研究绞股蓝皂苷对抗氧化酶活性的影响.方法 原代培养并建立人衰老皮肤成纤维细胞系,0、5、10、15 mg/L绞股蓝皂苷处理细胞,24、72、96 h后,四甲基偶氮唑盐(MTr)比色法检测细胞增殖;72 h后检测超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)活性.结果 药物作用24、72 h后,10、15 mg/L绞股蓝皂苷可以促进人衰老成纤维细胞增殖,增殖效应呈时间和浓度依赖(P<0.05).与0 mg/L比较,10、15 mg/L绞股蓝皂苷可以增强细胞SOD、CAT、GSH-Px活性(P<0.05),且与10 mg/L绞股蓝皂苷比较,15 mg/L绞股蓝皂苷能够显著提高SOD和GSH-Px活性(P<0.05).结论 绞股蓝皂苷可以通过提高SOD、CAT、GSH-Px活性,削弱氧化应激反应对细胞的损伤,使细胞增殖能力增强,延缓细胞衰老.

Abstract:Objective To study the effect of gypenosides (Gyp) on the activity of antioxidative enzymes at cellular level.Methods After the primary culture and subculture,human aged skin fibroblasts were incubated with Gyp (0,5,10,15 mg/L) for 24,72 and 96 h respectively.The MTT method was used to evaluate the cell proliferation and assay the bio logic activities of Gyp in different time and different doses.Relative level of the activity of SOD,CAT and GSH-Px in cells were measured by cytochemistry.Results Aged fibroblasts proliferation could be promoted by Gyp (10,15 mg/L) for 24,72 hours treatment (P 〈 0.05).The activity of SOD,CAT and GSH-Px increased in both group 10 and 15 mg/L Gyp as compared with group 0 mg/L (P 〈 0.05).Compared with 10 mg/L Gyp group,15 mg/L Gyp group showed significantly increased in the activity of SOD and GSH-Px (P 〈 0.05).Conclusion Gyp protects the aged fibroblasts by weakening oxidative stress,increasing the activity of antioxidative enzymes,and therefore delay the cells ageing.
