摘    要:目的:探讨利用组织工程技术,用自体血管壁组织培养预制血管的可行性。方法:取杂种犬12只,各切取右侧颈动脉2cm,将血管腔内残留血液冲洗干净,去除外膜纤维组织,用锐器切割和粉碎至匀浆状态,均匀种植于口径为3mm、长40mm的无纺聚羟基乙酸(polyglycolic acid,PGA)管状支架上;埋植于实验犬的腹直肌内。3月后,取出标本进行大体、组织学和超微结构观察。结果:PGA血管支架已完全被吸收和降解,种植的血管组织已完全生长成酷似血管的管状结构。电镜观察发现:外膜较薄,为疏松结缔组织;中膜较厚,主要由成纤维细胞、平滑肌细胞和胶原纤维等成分组成;内膜最薄,可见内皮细胞覆盖。结论:利用组织工程技术,将犬的动脉组织匀浆种植于PGA支架上,3月后支架完全降解和吸收,形成酷似血管的管状结构。

Abstract:ve:To study the feasibility of fabricating "blood vessels" from autologous arteries by wayof the tissue engineering technique. Methods: In a total of 12 dogs, a segment of the right commoncarotid artery, about 2cm in length, was obtained; the segment was rid of its adventitia, minced and sti-rred, thereby creating a tissue suspension, which was then evenly spreed on a tubular polyglycolic(PGA) scaffold 3mm in diameter and 40mm in length. The piece was imbedded into the rectus abdominismuscle of the dog. The grafts so prepared were exp1anted 3 months after implantation, and gross andhistological examination, as wel1 as the u1trastructure studies were performed. Results: The tubularPGA scaffolds were found to be completely degraded and absorbed, and the implanted vascular cellshad pro1iferated to take the form of a tubular structure resembling a natural artery. Scanning electronmicroscopical observation revealed that there was a new thin adventitia consisting of loose connectivetissue, the media was much thicker formed by smooth muscle cells and fibroblasts, and the neointimarepresented the thinnest layer made of endothelial cells. Conclusions: The canine carotid artery tissuesimplanted on a PGA scaffold could proliferate and form a tubular structure as the blood vessel and thePGA was completely degraded and absorbed 3 months after implantation.
