阿司匹林对大鼠局灶性脑缺血再灌注损伤中血NO、ET、TNF含量的影响

摘    要:目的探讨阿司匹林对大鼠局灶性脑缺血再灌注损伤中血一氧化氮(NO)、内皮素(ET)、肿瘤坏死因子(TNF)含量的影响.方法将实验动物随机分为阿司匹林组、模型组、假手术组、对照组.阿司匹林组予阿司匹林连续灌胃、其他3组予生理盐水连续灌胃20天后,采用颈内动脉线栓法造成大鼠大脑中动脉缺血再灌注模型,应用荧光分光光度法和放射免疫分析法观察4组血NO、ET、TNF含量的变化.结果假手术组血NO、ET、TNF与对照组比较无显著性差异(P>0.05);与假手术组比较,模型组NO显著下降(P<0.01),ET、TNF明显升高(均P<0.05);与模型组比较,阿司匹林组NO明显升高 (P<0.05),ET、TNF明显降低(均P<0.05).结论阿司匹林对大鼠局灶性脑缺血再灌注损伤具有显著的保护作用,其机制可能与血N0含量升高及ET、TNF降低有关.

Abstract:Objective To study changes of NO,ET,TNF content in blood in rats with focal cerebral ischemia reperfusion injury treated by Aspirin.Methods Animals were randomly divided into four groups: Aspirin group, model group, sham operative group, and control group. After Aspirin group given Aspirin and other groups given physiological saline by gastrogavage for 20 days, the middle cerebral artery (MCA) of rats was used thread embolism method of intenal carotid artery(ICA) to establish the rat models of focal cerebral ischemia reperfusion. Fluorospectrophotometry and radioimmunoassay were used to measure the changes of NO,ET,TNF in blood of four group.Results There was no obvious difference(NO,ET,TNF) between sham operative group and control group( P >0.05). Compared with the sham operative group, NO content in model group was obviously lower( P <0.01), ET and TNF content were obviously higher( P <0.05). Compared with the model group, NO content in Asprin group was obviously higher( P <0.05), ET and TNF content were obviously lower( P <0.05).Conclusion Aspirin had remarkable protective effect in treating rats with focal cerebral ischemia reperfusion injury, which may be related to increasing NO content and decreasing ET,TNF content in blood.
