人肝细胞肝癌组织中CLDN1和CLDN4的表达差异及其作用分析

目的探讨紧密连接蛋白CLDN1和CLDN4在人肝细胞肝癌(human hepatocellular carcinoma,HCC)组织中的表达及其意义。方法郑州大学第一附属医院2010年至2011年手术切除的HCC组织标本60例和正常肝组织50例,采用免疫组织化学二步法分别检测CLDN1和CLDN4的表达。结果 CLDN1和CLDN4在HCC组织中的阳性表达率分别为70%(42/60)和25%(15/60),两者的表达与患者年龄、性别、血液AFP浓度无关(P>0.05),与肿瘤分化程度无关(P>0.05)。CLDN1和CLDN4在HCC中的表达具有相关性(r=0.533,P<0.05)。CLDN1在正常肝组织中无表达(0/50),CLDN4在正常肝组织中高表达(50/50)。两者在正常肝组织中的表达具有明显差异(P<0.05)。结论肝细胞肝癌中存在CLDN1和CLDN4的异常表达,CLDN1在HCC中表达上调,而CLDN4在HCC中表达下调,两者表达具有相关性(r=0.533,P<0.05)。推测CLDN1表达上调和CLDN4表达下调能够促进肝细胞肝癌的发生发展。

Expression and clinical significance of CLDN1 and CLDN4 in human hepatocellular carcinoma

Objective To study the expressions and clinical significances of CLDN1 and CLDN4 in human hepatocellular carcinoma.Methods Immunohistochemical method was used to detect the expressions of CLDN1 and CLDN4 in both hepatocellular carcinoma tissues and normal liver tissues(HCC 60 cases and control group 50 cases)and the relation between CLDN1 and CLDN4 was evaluated.Results The positive rates of CLDN1 and CLDN4 expressions in HCC tissues were 70%(42/60) and 25%(15/60) respectively.The positive rates of CLDN1 and CLDN4 expressions in normal liver were 0(0/50) and 100%(50/50).The expressions of CLDN1 and CLDN4 have no relationships with the age,sex,AFP and degree of tumor differentiation of the patients with HCC.The expressions of CLDN4 had correlation with the expressions of CLDN1 in HCC.Conclusions The expressions of CLDN1 and CLDN4 in HCC are both abnormal.The expressions of CLDN4 in HCC are down regulated and the expressions of CLDN1 in HCC are over expressed.Abnormal expressions of CLDN1 and CLDN4 in HCC may promote the development and progression of HCC.
