海参糖胺聚糖对内皮细胞vWF和PAI-1表达的影响

摘    要:目的探讨玉足海参糖胺聚糖(GAG)对活化人脐静脉内皮细胞(HUVECs)von Willebrand因子(vWF)和纤溶酶原激活剂抑制物-1(PAI-1)表达的影响,以进一步明确GAG抗血栓作用机制。方法不同浓度GAG与凝血酶活化HUVECs共同孵育,MTT比色法检测GAG对细胞活力的影响,分别采用ELISA法和RQ—PCR法检测GAG对活化HUVECs vWF和PAI-1抗原蛋白和基因表达的影响。结果GAG可以降低活化HUVECsvWF抗原和PAI-1抗原的表达,5mg/LGAG抑制作用最为显著(P〈0.001),且强于相同剂量的肝素。5mg/LGAG还可以抑制活化HUVECsvWFmRNA和PAI-1 mRNA转录(P〈0.01、0.05),5mg/L肝素虽然对此两种mRNA转录亦呈抑制作用,但对PAI-1mRNA转录的抑制作用无统计学意义(P〉0.05)。结论GAG可能通过降低活化HUVECsvWF和PAI-1抗原表达及其mRNA转录,从而达到抗凝血及抗血栓作用。
