骨桥蛋白、基质金属蛋白酶-9和环氧化酶-2与脑膜瘤侵袭性的相关研究

摘    要:目的探讨骨桥蛋白(OPN)、基质金属蛋白酶-9(MMP-9)和环氧化酶-2(COX-2)在人脑膜瘤中的表达及其临床意义。方法采用酶联免疫吸附法检测40例脑膜瘤(侵袭性脑膜瘤16例,非侵袭性24例)患者和10例健康人血清中OPN、MMP-9和COX-2的浓度;采用免疫组化技术检测上述40例脑膜瘤组织及10例正常脑膜组织中OPN、MMP-9和COX-2的表达。结果健康人血清中OPN、MMP-9和COX-2浓度为0,正常脑膜组织中OPN、MMP-9和COX-2表达阴性。侵袭组中OPN、MMP-9、COX-2的血清浓度显著高于非侵袭组(P〈0.01)。OPN、MMP-9及COX-2蛋白在侵袭组中的表达均显著高于非侵袭组(P〈0.01)。在侵袭性脑膜瘤中MMP-9、OPN和COX-2三者的表达互为正相关(P〈0.01)。结论OPN、MMP-9、COX-2的血清浓度升高和在脑膜瘤组织中的表达上调与脑膜瘤的侵袭性密切相关,提示其可作为评价脑膜瘤侵袭性及预后的辅助指标。

Abstract:Objective To investigate the expressions of osteopontin (OPN), matrix metalloproteinase-9(MMP-9) and cyclooxygenase-2(COX-2) in meningiomas and their clinical meanings. Methods The samples of meningiomas were obtained from 40 patients, of whom, 16 had invasive meningiomas and 24 non-invasive ones. The serum concentrations of OPN, MMP-9 and COX-2 were detected by enzyme linked immunosorbent assay in all the patients with meningiomas and 10 healthful subjects. The expressions of OPN, MMP-9 and COX-2 were determined by immunohistochemical technique in all the meaningiomas and 10 samples of normal dural mater tissues, which were derived from the patients with cerebral trauma. Results The serum concentration of OPN, MMP-9, COX-2 in the healthful subjects and their expressions levels in the normal dural mater tissues were zero. The serum concentrations of OPN, MMP-9, COX-2 in the patients with invasive meningiomas were significantly higher than those in the ones with non-invasive meningiomas (P〈 0.01). The levels of OPN, MMP-9 and COX-2 expressions in the invasive meningiomas were significantly higher than those in the non-invasive ones (P〈0.01). Conclusion It is suggested that the serum concentration and expressions levels of OPN, MMP-9 and COX-2 in the meningiomas are closely correlated to the meningioma invasiveness.
