子宫内膜上皮内瘤变中PI3K和CyclinD1蛋白的表达及意义

摘    要:目的:探讨PI3K和CyclinD1蛋白表达在良性子宫内膜增生、子宫内膜上皮内瘤变(EIN)、子宫内膜样腺癌发生、发展中的作用。方法:选取增殖期子宫内膜10例、良性增生性子宫内膜30例(包括单纯性增生和复杂性增生),子宫内膜上皮内瘤变30例,子宫内膜样腺癌20例,应用免疫组织化学法检测各组中PI3K和CyclinD1蛋白的表达,分析其表达水平的变化。结果:PI3K和CyclinD1在增殖期子宫内膜、良性增生性子宫内膜、EIN和子宫内膜样腺癌中表达依次增高,组间差异具有统计学意义(H=46.64,P<0.01;H=44.96,P<0.01);PI3K与CyclinD1表达呈正相关(r=0.682,P<0.01),且二者与组别及年龄均呈正相关(P<0.01)。结论:PI3K高表达可能是通过诱导和调控CyclinD1的过表达实现的,PI3K和CyclinD1可能参与了子宫内膜恶性转化的过程。

Abstract:Objective:To explore the roles of PI3K and CyclinD1 protein in occurrence and development of benign endometrial hyperplasia,endometrial intraepithelial neoplasia(EIN),and endometrioid adenocarcinoma. Methods:Ten endometrium samples during proliferative phase,thirty benign proliferative endometrium samples(including simple proliferation and complex proliferation),thirty endometrium samples of EIN,and twenty endometrium samples of endometrioid adenocarcinoma were selected,immunohistochemical method was used to detect the expression levels of PI3K and CyclinD1 protein in different groups,and the changes of expression levels of PI3K and CyclinD1 protein were analyzed. Results:The expression levels of PI3K and CyclinD1 protein in endometrium during proliferative phase,benign proliferative endometrium,EIN,and endometrium of endometrioid adenocarcinoma increased gradually,there was statistically significant difference(H=46.64,P<0.01;H=44.96,P<0.01);there was a positive correlation between PI3K expression and CyclinD1 protein expression(γ=0.682,P<0.01),the expression levels of PI3K and CyclinD1 protein were positively correlated with group and age(P<0.01). Conclusion:High expression of PI3K may be realized by inducing and regulating over-expression of CyclinD1 protein,PI3K and CyclinD1 protein may participate in the course of malignant transformation of endometrium.
