国产盐酸加替沙星对细菌感染小鼠败血症的体内抗菌作用

摘    要:研究国产盐酸加替沙星(gatifloxacin)与氧氟沙星(ofloxacin)对细菌感染小鼠败血症的体内抗菌作用。 盐酸加替沙星与氧氟沙星对临床分离金葡球菌、肺炎双球菌、肺炎克雷伯氏菌和铜绿假单胞菌感染小鼠败血症的体内保护试验结果显示,盐酸加替沙星体内抗菌作用明显优于氧氟沙星,尤其是对肺炎双球菌、肺炎克雷伯氏菌的抗菌作用更显著优于氧氟沙星。比较试验两药ED50及ED95值结果,盐酸加替沙星和氧氟沙星对两株金葡球菌感染小鼠的体内保护作用ED50值及ED95值分别为3.77、3.94与11.17、12.54及8.86、8.25与29.18、36.30mg/kg(两药ED50值与ED95值的P值<0.01,具有显著差异),氧氟沙星的ED50值及ED95值比盐酸加替沙星大2.95~3.2倍及3.3~4.4倍。盐酸加替沙星和氧氟沙星对两株肺炎双球菌感染小鼠的体内保护作用ED50值及ED95值分别为2.89、3.38与20.47、23.36及8.82、8.06与62.10、62.45mg/kg(两药ED50值与ED95值的P值<0.01,具有显著差异),氧氟沙星ED50及ED95值比盐酸加替沙星大6.9倍及7.1~7.7倍。盐酸加替沙星和氧氟沙星对两株肺炎克雷伯氏菌感染小鼠的体内保护作用ED50值及ED95值分别为2.79、3.39与16.16、24.42及9.07、8.06与41.48、74.83mg/kg(两药ED50值和ED95值的P值<0.01,具有显著差异)。ED50值及ED95值氧氟沙星比盐酸加替沙星大5.8~6.3倍及4.6~9.24倍。盐酸加替沙星和氧氟沙星对两株铜绿假单胞菌感染小鼠的体内保护作用ED50及ED95值分别为21.88、37.49与54.02、73.81及140.73、118.12与199.5、335.88mg/kg(两药ED50值、ED95值的P值<0.01,具有显著差异),氧氟沙星的ED50及ED95值比盐酸加替沙星大1.97~2.47倍及1.42~2.84倍。

Abstract:In this paper we reported the study results of in vivo antibacterial activity of gatifloxacin and ofloxacin against the mouse septicemia infection by clinical isolates. The results showed that the antibacterial activity of gatifloxacin was more potent than that of ofloxacin when tested against S.aureus, S.pneumoniae, K.pneumoniae , and P.aeruginosa . Especially, gatifloxacin had evidently in vivo antibacterial activities against S.pneumoniae and K.pneumoniae , and was more potent than that of ofloxacin. The protection test showed that ED 50 and ED 95 values of gatifloxacin gaginst the mouse septicemia infected by S.aureus and P.aeruginosa were 2.95~3.2, 3.3~3.4 and 1.97~2.47, 1.42~2.84 fold lower than that of ofloxacin. The ED 50 and ED 95 values of gatifloxacin against the mouse septicemia infectied by S.pneumoniae and K.pneumoniae were 6.9, 7.1~7.7 and 5.86.3, 4.6~9.24 fold lower than that of ofloxacin.
