阿奇霉素对人肺腺癌多药耐药细胞株A549/ADM的耐药逆转研究

摘    要:目的研究阿奇霉素(AZM)对人肺腺癌多药耐药细胞株A549/阿霉素(ADM)的耐药逆转作用。方法使用四甲基偶氮唑盐(MTT)法检测不同浓度AZM对细胞株A549/ADM的抑制率,确定其非细胞毒性浓度。用MTT法分别检测ADM、顺铂(DDP)、长春新碱(VCR)、氨甲蝶呤(MTX)对细胞株A549、A549/ADM以及使用AZM处理后的A549/ADM的半数抑制浓度(IC50)。使用流式细胞仪检测A549以及加入AZM前后的A549/ADM内ADM的荧光强度。结果≤10mg/L的AZM对A549/ADM无明显细胞毒作用。A549/ADM对ADM、VCR、DDP、MTX均有不同程度的耐药。使用5mg/LAZM处理后,A549/ADM对上述4种药物的敏感度均有不同程度的提高。AZM的浓度提高至10mg/L时,A549/ADM对上述药物的敏感程度提高更明显。A549/ADM经AZM处理后细胞内ADM荧光强度明显提高。结论AZM可以部分逆转A549/ADM的耐药,其逆转效果与剂量相关。

Abstract:Objective To investigate the reversal effect of azithromycin on human lung adenocarcinoma multidrug resistance cell line A549/adriamycin(ADM).Methods The non-cytotoxic concentration of azithromycin(AZM) to A549/ADM was determined with MTT assay. The IC_ 50 of ADM,DDP,VCR and MTX to A549,A549/ADM and A549/ADM treated with AZM were detected with MTT assay. The fluorescence intensity of ADM in A549 and A549/ADM with or without AZM treated were detected with flow cytometry (FCM). Results No more than 10 mg/L AZM has no obvious cytotoxicity to A549/ADM cell line. A549/ADM to ADM,DDP, VCR, MTX were drug resistance incoordinately than A549. With AZM added(5 mg/L), sensitivities of A549/ADM to ADM, DDP,VCR,MTX all increased incoordinately. The sensitivities of A549/ADM to these drugs were much higher when treated with 10mg/L AZM. The fluorescence intensity of ADM in A549/ADM treated with AZM increased obviously. Conclusion AZM could partially reverse the drug resistance of A549/ADM and the effect of reversal correlates with concentrations.
