生脉散对急性肝衰竭大鼠内毒素诱导细胞因子水平的影响

摘    要:目的探讨生脉散在不同时间点对急性肝衰竭大鼠内毒素(脂多糖,LPS)诱导细胞因子水平的影响。方法D-氨基半乳糖腹腔注射建立大鼠急性肝衰竭模型,拟用LPS攻击建立肝衰竭内毒素血症模型。用生脉散治疗与对照组比较。取造模后2h及8h两个时间点,检测大鼠血清LPS与细胞因子如肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)、白介素-6(IL-6)、细胞间黏附分子-1(ICAM—1)、一氧化氮(NO)、诱导型一氧化氮舍酶(iNOS)和总一氧化氮合酶(zNOS)水平。结果2h后生脉散可减低急性肝衰竭内毒素血症大鼠血清LPS、TNF—α、IL-1β、IL-6、ICAM-1和NO水平,差异有统计学意义(P〈0.05);8h后生脉散可减低急性肝衰竭内毒素血症大鼠血清LPS、TNF—α、IL-1β和ICAM—1水平,差异有统计学意义(P〈0.001)。结论急性肝衰竭状态下,可存在不同程度的内毒素血症,并引起细胞因子水平变化的炎症级联反应,生脉散通过调节细胞因子水平起到治疗作用。

Abstract:Objective To study the effects cell factors induced by lipopolysaccharide in rats of Shengmai decoction (SMD) on the level of with acute liver failure. Methods The models of acute liver failure were induced by D-galactoasmine (D-Gal), then, based on which endotoxin models by LPS, and Shengmai decoction therapy was compared with control group. At the points of 2nd and 8th hours after modeling, the serum level of Lipopolysaccharide (LPS), TNF-α, IL-1β, IL-6, ICAM-1, No, iNOS and zNOS were detected. Results After 2 hours, SMD could significantly reduce the level Of LPS, TNF-α, IL-1β, IL-6, ICAM-1 and No in rats with acute liver failure induced by LPS (P〈0.05); and after 8 hours, SMD can significantly reduce the level of LPS, TNF-α, IL-1β, IL-6 and ICAM-1 in the rats (P〈0.001). Conclusion In the SD rats with acute liver failure, the different levels of serum endotoxin are existed and the level of cell factors diversification inflammation progressional reactions can be induced, and SMD can regulate the level of cell factors to treat the disease.
