摘    要:目的:研究白杨素(ChR)是否具有增敏重组人可溶性肿瘤坏死因子相关凋亡诱导配体(rhsTRAIL)诱导人肝癌Bel-7402细胞凋亡作用。方法:体外培养人肝癌Bel-7402细胞。碘化丙啶(PI)染色流式细胞术(FCM)分析细胞死亡率。DNA琼脂糖凝胶电泳确证诱导细胞凋亡作用。Western Bloting检测细胞DR5蛋白的表达。结果:ChR40μmol/L、rhsTRAIL 100ng/mL以及两者合用的细胞死亡率分别为4.91%±0.38%、5.89%±0.39%和28.7%±2.50%。ChR(40μmol/L)联合rhsTRAIL(100ng/mL)处理48小时,人肝癌Bel-7402细胞展示出典型DNA梯形条带图谱。Western Blot分析结果发现:白杨素以浓度和时间依赖的方式上调Bel-7402细胞DR5表达。结论:亚细胞毒性浓度的白杨素具有敏化rhsTRAIL诱导人肝癌Bel-7402细胞凋亡作用。

Abstract:Objective o investigate whether chrysin(ChR) sensitizes apoptosis induced by recombinant human solubility TNF-related apoptosis-inducing ligand(rhsTRAIL) in human hepatocellular carcinoma(HCC) Bel-7402 cell line.Methods Bel-7402 cell line was cultured in vitro.The cell death rate was determined by flow cytometry using PI fluorescence staining.The characteristic features of cell apoptosis was examined using DNA agarose gel electrophoresis.The expressions of DR5 were analyzed by Western blot.Results The cell death rate in human hepatocellular carcinoma Bel-7402 cell line by ChR(40μmol/L) or rhsTRAIL(100ng/mL) or both for 48h were 4.91%±0.38%,5.89%±0.39% and 28.7%±2.50%,respectively.The ladder band could be shown in DNA agarose gel electrophoresis after treatment with rhsTRAIL(100ng/mL) for 48h pretreated with ChR(40μmol/L) for 30 minute.ChR upregulated expression of DR5 in Bel-7402 cells,in a time-and concentration-dependent manner.Conclusion ChR at suboptimal concentration possess sensitization of rhsTRAIL induced apoptosis of human hepatocellular carcinoma Bel-7402 cells.
