反义c-myc寡核苷酸对大鼠自体移植静脉内膜增生的影响

目的 研究反义ｃ－ｍｙｃ寡聚脱氧核苷酸（ＯＤＮ）对大鼠自体移植静脉内膜增生的影响。方法 移植静脉外周涂以加有反义ｃ－ｍｙｃ寡聚脱氧核苷酸的Ｆ１２７凝胶，于术后１、２周测量静脉内膜平均厚度及观察血管内膜内ｃ－ｍｙｃ蛋白表达情况。结果 手术后１、２周，移植静脉内膜的平均厚度、内膜中ｃ－ｍｙｃ蛋白表达在反义ｃ－ｍｙｃ寡聚脱氧核苷酸组均减少，并存在量效关系（Ｐ〈０．０１）。而只用Ｆ１２７胶组及正义寡聚脱氧

Effect of antisense c-myc oligodeoxynucleotides on intimal hyperplasia of vein autografts of rats

Objective To study the inhibitory effect of antisense c-myc oligodeoxynucleotides on intimal hyperplasia of vein autografts. Methods Antisense c-myc oligodeoxynucleotides of different concentrations, which was dissolved in 30 % pluronic F127 gel solution, was applied around the transplanted vein adventitia. The samples were got 1 and 2 weeks after operation to measure the thickness and observe the expression of c-myc protein in intima. Results At the first week and second week postoperatively, the thickness of vein intima and the c-myc protein expression index were obviously inhibited in a dose-dependent manner in the antisense group as compared with that in the control groups ( P < 0.01). But in the groups of F127 or sense oligodeoxynucleotides only no inhibitory effects were found. Conclusions Antisense c-myc oligodeoxynucleotides can inhibit intimal hyperplasia of rat vein autografts in a dose-dependent manner, which was contributed to the decrease of the expression of c-myc protein in intima.;
