白芍总苷对心肌缺血再灌注模型大鼠心肌细胞凋亡的影响

摘    要:目的 :观察白芍总苷对心肌缺血再灌注模型大鼠心肌细胞凋亡的影响。方法 :制备大鼠心肌细胞缺血再灌注模型,随机分为假手术组、缺血再灌注模型组及低、中、高剂量白芍总苷组,比较各组心肌细胞存活率;检测乳酸脱氢酶(LDH)释放;流式细胞术检测细胞凋亡;免疫印迹法检测心肌组织Wnt3a、磷酸化GSK-3β(p-GSK-3β)和β-catenin的表达水平。结果 :与缺血再灌注模型组相比,白芍总苷不同浓度各组心肌细胞Wnt3a、p-GSK-3β、β-catenin蛋白表达、LDH释放及细胞凋亡率均显著降低,且白芍总苷呈浓度依赖性效应。结论 :白芍总苷对心肌缺血再灌注模型大鼠心肌细胞凋亡具有保护性抑制作用,其机制可能与白芍总苷能抑制Wnt3a表达,从而影响Wnt/β-catenin信号通路密切相关。

Abstract:Objective:To investigate the effects of total glucosides of paeonia(TGP)on cardiomyocyte apoptosis of myocardial ischemia in rats.Method:Cultured neonatal rat cardiomyocytes(NRCs)were subjected to simulate ischemia/reperfusion(I/R).NRCs were randomly divided into 5 groups:control group,IR group,TGP low-dose group(1μmol/L),TGP middle-dose group,and TGP high-dose group.NRC injury was determined by MTT and lactate dehydrogenase(LDH)leakage assay.Apoptosis was assessed by flow cytometry.The level of myocardial Wnt3a,GSK-3β(p-GSK-3β),andβ-catenin expression was measured by Western blotting.Results:Compared with IR group,the expressions of Wnt3a,GSK-3β(p-GSK-3β),β-catenin,apoptosis and LDH leakage were lower in the TGP group than those in I/R group.Conclusion:The protective effect of TGP on cardiomyocyte apoptosis of myocardial ischemia reperfusion is obvious and Wnt/β-catenin signaling pathway may be involved in the mechanism.
