摘    要:目的:研究过氧化物酶6(Prdx6)在慢性砷中毒大鼠附睾中的表达及意义,为砷中毒导致的男性不育提供理论依据.方法:健康清洁级雄性SD大鼠随机分为高(60 mg/L)、中(12 mg/L)、低(2.4 mg/L)剂量砷染毒组和对照(蒸馏水)组,采用经口自由饮用方式进行染毒,连续染毒6个月.采用免疫印迹及实时荧光定量PCR法检测Prdx6的表达变化及意义,透射电镜观察精子超微结构变化.结果:免疫印迹及实时荧光定量PCR结果显示,与对照组比较,各染毒组Prdx6的蛋白及mRNA表达水平均显著较低.透射电镜结果显示对照组精子结构正常,砷染毒各组精子尾部中段线粒体均出现了病理损害.结论:慢性砷中毒对大鼠附睾中Prdx6的表达有影响,引起精子超微结构改变,说明其与男性不育有一定的关系.

Abstract:Objective :To study the effect of chronic arsenic poisoning on Peroxiredoxins 6(Prdx6) expression in the epididymis of rats and its effect on the structure of sperms,and to provide theoretical basis for male infertility caused by arsenic poisoning. Methods :Forty male Sprague-Dawley rats were randomly divided into high-dose arsenic infected group (60.0 mg/L),middle-dose arsenic infected group (12.0 mg/L),low-dose arsenic infected group (2.4 mg/L) and control group(distilled water group). Rats were treated with arsenism through drinking water for 6 consecutive months.At the end of the experiment,the protein and mRNA expression levels of Prdx6 were observed by Western blotting and RT-PCR. The ultrastructure of sperms was observed by transmission electron microscopy. Results :Results of Western blotting and RT- PCR showed that compared with control group,the protein and mRNA expression levels of Prdx6 in each infected group were lower,and the differences were statistically significant. The results of transmission electron microscopy showed that the sperm structure of the control group was normal,and the mitochondria of the middle part of the sperm tail of each infected group showed pathological damage. Conclusion : Arsenism affects the expression of Prdx6 in the epididymis and causes ultrastructural damage,which indicates that it may play an important role in male infertility.
