响应面法优化土人参根中总黄酮提取及其抗氧化活性研究

摘    要:目的 采用超声波辅助提取苗药用的土人参根中总黄酮,比较不同提法所得总黄酮的抗氧化活性。方法 在单因素试验基础上采用响应面法优化超声波处理后固液比、乙醇体积分数、浸提温度及其交互作用对总黄酮得率的影响,DPPH.、超氧阴离子与羟基自由基清除法考查水浴、超声波辅助和酶辅助提取的苗药土人参根中总黄酮的抗氧化活性。结果 苗药用土人参根中总黄酮的最佳工艺为固液比1:26、乙醇体积分数65%、浸提温度70℃,在此条件下,总黄酮得率为 2.62% ,与回归方程预测值 2.603% 符合良好。结论 该优化工艺实现了土人参根中黄酮类化合物的高效提取,所得总黄酮以酶法提取的抗氧化性能力最强。目的 采用超声波辅助提取苗药用的土人参根中总黄酮,比较不同提取方法所得总黄酮的抗氧化活性。方法 在单因素试验基础上采用响应面法优化超声波处理后固液比、乙醇体积分数、浸提温度及其交互作用对总黄酮得率的影响,DPPH?、超氧阴离子与羟基自由基清除法考查水浴、超声波辅助和酶辅助提取的苗药土人参根中总黄酮的抗氧化活性。结果 苗药用土人参根中总黄酮的最佳工艺为固液比1:26、乙醇体积分数65%、浸提温度70℃,在此条件下,总黄酮得率为 2.620% ,与回归方程预测值 2.603% 符合良好。三种提取方法所得总黄酮的抗氧化活性以酶法提取的最强且均好于Vc。结论 该优化工艺实现了土人参根中黄酮类化合物的高效提取,不同提取方法得到的总黄酮抗氧化活性差异明显。

Abstract:OBJECTIVE The ultrasonic-assisted extraction method was used for the extraction of flavonoids from the Talinnum paniculatum and its antioxidant activity in vitro was compared . METHODS On the basis of the single factors, the effects of liquid-solid ratio, ethanol volume fraction and their interaction for extraction of flavonoids has been investigated using Box-Behnken Design response surface method. The antioxidant activities of three methods used to extract the flavoniods: ultrasonic-assisted, water bath and enzymic methods were compared by the experiments: scavenging capabilities of DPPH?free radical, hydroxyl free radical and superoxide aninoic free radical. CONCLUSION The optimal extraction conditions were as follows: liquid-solid ratio 26:1, ethanol volume fraction 62% and extraction time 3h . Under the above conditions, the extraction yield of flavonoids were 2.620%, which is in accord with the predicted values 2.603%. The total flavonoids extracted by the above three methods and the antioxidant activities were compared. As a result, total flavonoids of Talinnum paniculatum has the better antioxidant activity than Vc and the enzymic methed has the best antioxidant activity. CONCLUSION The target of extracting flavonoids compound efficiently from Talinum has been achieved by the optimized technology, The flavonids extracted using different methods has the significantly different antioxidant activity.
