中波紫外线辐射刺激HaCaT细胞产生IL-6及法莫替T的抑制作用

摘    要:目的:观察中波紫外线辐射对体外培养的HaCaT细胞产生IL-6的影响及法莫替丁的作用。方法:用不同剂量中波紫外线辐射培养的HaCaT细胞,加入不同浓度的法莫替丁(分别为0、10-7、10-6、10-5、10-4mol/L),在不同时点收集上清液,ELISA法检测IL-6浓度。结果:与未辐射组相比,HaCaT细胞经300、600J/m2剂量的中波紫外线辐射后4h IL-分泌开始显著增加,经100J/m2剂量的中波露外线辐射后24h IL-6分泌开始显著增加(P<0.05);与未加法莫替丁组相比,HaCaT细胞经300J/m2辐射加入法莫替丁后12、24h可显著抑制IL-6的产生(P<0.05),最大抑制率为51.85%,出现在加入法莫替丁24h浓度为10-4mol/L时。结论:中波紫外线辐射可刺激HaCaT细胞产生IL-6,法莫替丁可抑制这种作用。

Abstract:Objective: To study the effect of ultraviolet B radiation on the release of IL-6 in HaCaT cell line and the inhibition of famotidine on this effect. Methods: HaCaT keratinocytes were incubated without or with famotidine (concentration 10-7 - 10-4 mol/L) under various ultraviolet B treatment modalities in vitro, supernatants of the cell cultures collected at different time points and IL-6 detected by ELJSA kit. Results: Compared with no radiation, an increase of IL-6 in vitro upon ultraviolet B radiation from HaCaT keratinocytes was observed from 4 h for 300 J/m2 , from 4h for 600 J/m2, and from 24 h for 100 J/m2, respectively ( P < 0. 05). The production of IL-6 induced by ultraviolet B radiation from HaCaT keratinocytes was inhibited at 12 h and 24 h after famotidine was added into cultures compared with no famotidine (P < 0. 05), the inhibitory maximum of IL-6 being 51. 85% at 24 h for 10"1 mol/L famotidine. Conclusions: It is suggested that ultraviolet B radiation can induce the release of IL-6 in HaCaT cell line and famotidine can inhibit this effect.
